Overview

Description

Connexin-43, a member of the alpha-type (group II) subfamily in the connexin family, acts as an integral membrane protein. Through the formation of hexamers, connexin-43 assembles connexons that cluster together to create gap junctions. These junctions allow the diffusion of low molecular weight substances from one cell to an adjacent cell. Additionally, connexin-43 is believed to have a crucial role in the physiology of hearing, specifically in the potassium recycling process within the cochlear endolymph. By participating in this recycling mechanism, connexin-43 contributes to the maintenance of proper auditory function.

Other names

Connexin 43, also known as Connexin-43, Cx 43, Cx43, CXA1_HUMAN, DFNB38, is a vital protein involved in heart function. It is classified as a 43 kDa Gap junction protein and plays a crucial role in cell-to-cell communication. This protein is responsible for the formation of gap junctions, which are channels that allow the exchange of ions and small molecules between adjacent cells.
In addition to its role in the heart, Connexin 43 has been linked to various genetic disorders. Mutations in the GJA1 gene, which encodes this protein, have been associated with conditions such as DFNB38, Oculodentodigital dysplasia (ODDD), and Syndactyly type 3 (SDTY3).
The Gap junction alpha-1 protein, as it is sometimes referred to, is also known by other names like Gap junction protein alpha 1 43kDa (connexin 43), Gap junction protein alpha 1 43kDa, Gap junction protein alpha-like, GJA 1, Gja1, GJAL, ODD, ODDD, ODOD.
In summary, Connexin 43, or Cx43, is a crucial protein involved in heart function and cell-to-cell communication. Its various names and associations with genetic disorders highlight its significance in maintaining cellular connectivity and overall physiological processes.

Application

For Western blotting, the suggested dilution range is between 1:500 to 1:3000, while for immunohistochemistry, it is recommended to dilute the antibody in the range of 1:50 to 1:200. For fluorescence microscopy, the suggested dilution is between 1:100 to 1:500, and for ELISA using peptide, the suggested dilution range is between 1:20000 to 1:40000. These dilution ranges have been optimized to ensure reliable and reproducible results. Whether you are performing basic research or conducting advanced experiments, this antibody will provide a highly specific and reliable response for your scientific needs.

Immunogen

An artificial peptide has been produced from Connexin 43 found in humans. The newly formed peptide closely resembles the original protein sequence.

Background

A regulator of bladder capacity is a gap junction protein that is involved in various physiological processes. These gap junction proteins are composed of closely packed transmembrane channels called connexons. The primary function of these channels is to enable the diffusion of small molecules between neighboring cells. One of the significant roles of this protein is its involvement in the recycling of potassium in the cochlear endolymph, which is crucial for hearing.
Interestingly, this protein also acts as a negative regulator of bladder functional capacity. It achieves this by promoting intercellular communication through electrical and chemical signals, thereby enhancing the sensitivity of bladder muscles to cholinergic neural stimuli. Consequently, the muscles contract, leading to a decrease in bladder capacity. Additionally, this protein is known to play a role in inhibiting cell growth by regulating the expression and localization of another protein called NOV.
Furthermore, it is worth noting that this protein is essential for gap junction communication in the ventricles of the heart. While the exact mechanisms may vary, this protein's role in maintaining efficient intercellular communication is vital for proper functioning of the ventricles.

Tissue specificity

Expressed in the heart and fetal cochlea.

Posttranslational modification

Phosphorylated at Ser-368 by PRKCG; phosphorylation induces disassembly of gap junction plaques and inhibition of gap junction activity (By similarity). Phosphorylation at Ser-325, Ser-328 and Ser-330 by CK1 modulates gap junction assembly. Phosphorylation at Ser-368 by PRKCD triggers its internalization into small vesicles leading to proteasome-mediated degradation (By similarity).Sumoylated with SUMO1, SUMO2 and SUMO3, which may regulate the level of functional Cx43 gap junctions at the plasma membrane. May be desumoylated by SENP1 or SENP2.S-nitrosylation at Cys-271 is enriched at the muscle endothelial gap junction in arteries, it augments channel permeability and may regulate of smooth muscle cell to endothelial cell communication.
