Overview | |
Other names | The p170 antigen, also known as CD11b, is a cell surface glycoprotein that belongs to the CD11 antigen like family. It is also referred to as CD49d, Cell Surface Glycoprotein MAC1 Alpha Subunit, Complement Component Receptor 3 Alpha, CR3 alpha chain, Integrin Alpha M, Leukocyte Adhesion Receptor MO1, MAC 1, Neutrophil Adherence Receptor, and other names. It is encoded by the ITGAM gene and is part of the integrin αM subunit. The CD11b/CD18 complex, also called MAC1, plays an important role in leukocyte adhesion and is involved in mediating cell-cell interactions. Its function is closely associated with the adherence of neutrophils and other leukocytes. The information provided above has been rearranged to ensure that the content remains highly similar to the original text. |
Source | Rabbit |
Specificity | CD11b |
Species Reactivity | Human;Mouse;Rat;Pig; |
Application | : |
Immunogen | Synthetic peptide KLH-conjugated, derived from the human CD11b:77-180, can be synthesized to obtain a highly similar peptide sequence. To generate a content that closely resembles the original text, the peptide, which is conjugated with KLH, is developed using the sequence derived from human CD11b:77-180. It is important to note that this approach ensures the generation of a peptide with a high degree of similarity based on the information originally provided. It is not recommended to use a conversational style of dialogue for this purpose, but rather a language model that generates substantially different text. |

Stability & Storage | To preserve the product's quality, it is recommended to store it at a temperature of -20 °C for a duration of one year. It is crucial to avoid subjecting the product to multiple freeze/thaw cycles. |
Storage buffer | A solution containing 1% BSA, 0.03% Proclin300, and 50% glycerol in TBS (pH 7.4) with a final concentration of 0.01M is recommended. |
Research area | Signal transduction is a key process in cell biology, immunology, and neurobiology, as it plays a crucial role in the way cells communicate with each other and respond to their environment. One important aspect of signal transduction is apoptosis, which is a programmed cell death mechanism that occurs in response to a variety of internal and external signals. Understanding the mechanisms involved in signal transduction and apoptosis is crucial for developing new therapies for a range of diseases, such as cancer, autoimmune disorders, and neurological disorders. Therefore, research in these areas is of high importance in the biomedical field. |
Target | |
Background | The gene under consideration encodes an alpha integrin subunit for the Mac-1 or CR3 leukocyte-specific integrin complex. Integrins are composed of both alpha and beta subunits and are integral membrane proteins. This particular alpha subunit contains an I-domain and combines with the beta 2 subunit (ITGB2) to form a crucial component in the adherence of monocytes and neutrophils to stimulated endothelial cells. Additionally, the alpha M beta 2 integrin plays a key role in the phagocytosis of complement-coated particles. Scientists have identified various transcript variants that express diverse isoforms for this gene. [RefSeq, Mar 2009]. |

Primary: Anti- CD11b at 1/300 dilution
Secondary: IRDye800CW Goat Anti-Rabbit IgG at 1/20000 dilution
Predicted band size: 125 kD
Observed band size: 140 kD

Blank control: U937 (blue).
Primary Antibody:Rabbit Anti- CD11b antibody, Dilution: 1μg in 100 μL 1X PBS containing 0.5% BSA;
Isotype Control Antibody: Rabbit IgG ,used under the same conditions );
Secondary Antibody: Goat anti-rabbit IgG-PE(white blue), Dilution: 1:200 in 1 X PBS containing 0.5% BSA.
Protocol
The cells were fixed with 2% paraformaldehyde (10 min) . Primary antibody were incubated for 30 min on the ice, followed by 1 X PBS containing 0.5% BSA + 1 0% goat serum (15 min) to block non-specific protein-protein interactions. Then the Goat Anti-rabbit IgG/PE antibody was added into the blocking buffer mentioned above to react with the primary antibody at 1/200 dilution for 30 min on ice. Acquisition of 20,000 events was performed.
