Background

This protein is a serine/threonine-protein kinase that is essential for proper chromosome alignment and spindle-assembly checkpoint signaling during mitosis. Its key role is to assemble checkpoint proteins at the kinetochore and localize proteins such as CENPF, BUB1B, CENPE, and MAD2L1. It also plays a crucial role in defining the localization of SGO1 and thereby affecting sister chromatid cohesion. Additionally, this protein is necessary for ensuring proper chromosome segregation and binding to BUB3 is required for this function. It can regulate chromosome segregation in a kinetochore-independent manner and phosphorylate BUB3.
The BUB1-BUB3 complex, of which this protein is a part, plays a vital role in inhibiting the activity of anaphase-promoting complex or cyclosome (APC/C) by phosphorylating its activator CDC20. Kinase activity is necessary for the inhibition of APC/CCDC20 and chromosome alignment, but it does not play a major role in spindle-assembly checkpoint activity. Furthermore, this protein mediates cell death in response to chromosome missegregation and acts to suppress spontaneous tumorigenesis. Tissue specificity

Posttranslational modification

Upon spindle-assembly checkpoint activation it is hyperphosphorylated and its kinase activity toward CDC20 is stimulated. Phosphorylation at Thr-609 is required for interaction with PLK1, phosphorylation at this site probably creates a binding site for the POLO-box domain of PLK1, thus enhancing the PLK1-BUB1 interaction.Ubiquitinated and degraded during mitotic exit by APC/C-Cdh1.
