Description | This enzyme is known as an alkyl DNA dioxygenase and is responsible for repairing DNA damage caused by alkylation, specifically targeting 1-methyladenine and 3-methylcytosine. It achieves this through a process called oxidative demethylation. Notably, this dioxygenase exhibits a strong preference for single-stranded DNA structures. Additionally, there is evidence to suggest that it may also act on RNA molecules. In order to function, this enzyme requires the presence of molecular oxygen, alpha-ketoglutarate, and iron. By utilizing these essential cofactors, the alkyl DNA dioxygenase can effectively carry out its demethylation repair mechanism. |
| Other names | ABH3, also known as ALKBH3 or DEPC1, is an alkylated DNA repair protein which belongs to the alpha-ketoglutarate-dependent dioxygenase alkB homolog family. It is a homolog of the E. coli alkylation repair protein alkB and plays a vital role in repairing DNA damage caused by different types of alkylating agents. The gene encoding ABH3 is located on human chromosome 16 and is expressed in a wide range of tissues. ABH3 has been implicated in the development of prostate cancer and is considered to be a potential marker for this disease. Besides, it has been shown to be involved in regulating RNA methylation and DNA demethylation, suggesting a broader role in epigenetic regulation. In summary, ABH3 serves as an essential component of the DNA repair machinery and has emerged as a promising target for cancer therapy. |
