Format

A concentrated solution in 20mM PB, 150mM NaCl, 1mM EDTA, pH7.4 was filtered through a 0.2μm filter and then lyophilized. To create a similar content, the filtered solution was first concentrated, before undergoing a filtration process. The solution was then lyophilized to create a dried state. The solution had a pH level of 7.4 and was made up of 20mM PB, 150mM NaCl, and 1mM EDTA.

Endotoxin_level

The level of Recombinant HumanIL-33 present in the sample was found to be less than 1 EU/μg, which was determined using the LAL method. It is essential to maintain a low concentration of this protein to ensure accurate experimental results.

Activity

The biological activity of the standard is equivalent to that of the fully active ED.₅₀The specific activity of less than 0.05 ng/ml, as determined by a cell proliferation assay using murine D10S cells, indicates that the measured substance has a highly potent effect. This potency is reflected in its ability to induce cell proliferation at a concentration higher than 2.0 × 10 in a given volume.⁷ IU/mg.

Reconstitution

To ensure the contents settle at the bottom, we suggest briefly centrifuging the vial before opening it. Reconstitute the vial by adding sterile distilled water or an aqueous buffer with 0.1% BSA, aiming for a concentration of 0.1-1.0 mg/mL. To maintain stability, divide the solution into working aliquots and store them at a temperature of no higher than -20°C. For further dilutions, use appropriate buffered solutions.

Stability & Storage

To ensure optimal stability of this freeze-dried preparation, it is recommended to store it at 2-8°C, although for long-term preservation, it is best to keep it at -20°C in a desiccated state. Once reconstituted, the preparation can remain stable for up to one week at 2-8°C. For maximum potency, it is advisable to divide the reconstituted preparation into working aliquots and store them in the temperature range of -20°C to -70°C. Avoid exposing the preparation to repeated freeze-thaw cycles, as this can adversely affect its stability.
