Description

Description

ACK Lysis Buffer, also referred to as Red Blood Cell Lysis Buffer, is designed to effectively lyse and eliminate nuclearless red blood cells from fresh blood or tissue samples of mammalian species including humans, rats, and mice. Its unique formula ensures minimal damage to lymphocytes and other nucleated cells while facilitating red blood cell lysis. By using this product, it is possible to obtain more than 80% of the total white blood cells in the sample while maintaining their cellular activity.

This product is primarily comprised of ammonium chloride, which is an ideal solution for quick and easy lysis of red blood cells, as well as efficient enrichment and separation of white blood cells. With a ready-to-use formula, this product offers a convenient and effective solution for cell isolation and sample preparation. In addition, this product is sterilized to ensure that isolated cell samples remain uncontaminated and are suitable for use in subsequent primary cell culture, cell fusion, flow sorting, nucleic acid or protein extraction, and a range of routine analyses and detection procedures. Overall, this product provides a reliable and efficient means of isolating and preparing high-quality cell samples for use in a wide range of research applications.

The principle of cell membrane burst to lysis of anucleated red blood cells is utilized by this product, which takes advantage of the difference in salt ion concentration inside and outside the cell. Following a sterile treatment, its primary application is in the separation and purification of dispersed and digested tissue cells. Moreover, it is also employed in the isolation and purification of lymphocytes, as well as the elimination of red blood cells during experiments involving tissue cell protein and nucleic acid extraction.

The lysate available for use is free of any contaminants and is considered a sterile product. It is important to be diligent in maintaining sterile operations when handling and separating cells for use in cell culture. Maintaining a sterile environment is critical to ensuring successful and accurate results in any cell culture process. Please ensure that you follow the proper protocols and guidelines to maintain a sterile environment and avoid any contamination in your experiments.

To prepare a blood sample for further testing, follow these steps. Start by adding three times the volume of erythrocyte lysate to the blood sample and mix it well. Leave the mixture at room temperature or 4ºC for about 5 minutes, making sure to mix it upside down a few times during this period. Then, centrifuge the mixture at 10000rpm for 1 minute. If the maximum centrifuge speed is not allowed, centrifuge it at 3000rpm for 5 minutes. Make sure to perform this step at 4ºC for better results. Remove the red supernatant after centrifugation. Repeat steps 2 and 3 once if incomplete lysis of red blood cells is found. Only a very small amount of red blood cells should remain to ensure accurate testing. Clean the sample with an appropriate buffer, such as PBS with pH between 7.2 and 7.4, one to two times. After centrifugation, the white blood cell precipitation should be left for subsequent operations.
