Description

The three-color pre-dyed protein standard is a reliable product that consists of 12 meticulously purified pre-dyed proteins. These proteins span a wide range of molecular weights, ranging from 10 to 245kD. In the tris-glycine buffer system, each protein band exhibits a distinct color. Specifically, the 25kD band appears green, the 75kD band appears red, while the remaining bands display a vibrant blue hue.
This protein standard serves a crucial purpose in Western blotting, where it aids in determining the success of membrane transfer and allows for the estimation of protein molecular weights. It is compatible with various types of membranes, such as PVDF, NC, and NYLON films.
To facilitate convenience and efficiency, the proteins have already been prepared in a loading buffer, eliminating the need for additional preparation steps. Each protein in the standard has a concentration ranging from 0.1 to 0.4 mg/mL.
For optimal storage and preservation, the protein standard is stored in a buffer consisting of 20 mM Tris-phosphate with a pH of 7.5 at a temperature of 25 degrees Celsius. The storage buffer further comprises 2% SDS, 0.2 mM Dithiothreitol, 3.6 M Urea, and 15% (V/V) Glycerol. These conditions ensure the stability and longevity of the protein standard.

Form
The buffer used contains 20 mM Tris-phosphate, with a pH of 7.5 at 25°C. Each protein is present in the buffer at a concentration of approximately 0.1-0.4 mg/ml. The buffer also contains 2% SDS, 0.2 mM Dithiothreitol, and 15% (v/v) Glycerol. In addition, the buffer is enriched with 3.6 M Urea to enhance protein stability and solubility. These conditions are ideal for protein analysis and facilitate the separation of proteins based on size and charge using gel electrophoresis.
Protocol
1. Before use, allow the product to melt by keeping it at room temperature.
2. Thoroughly mix the product and simultaneously test a sample for SDS-PAGE by taking 5μL of it.
3. Store any unused rainbow pre-dyeing marker following the specified storage conditions to maintain its quality.
Operating Instructions
Here are some possible rearranged versions:
- This product doesn't require any heating before use. To ensure accurate electrophoretic results, make sure to regularly replace the buffer and use a freshly prepared gel. The product can be kept at 4°C for up to 3 months, but if you need to store it longer, store it at -20°C. To prevent contamination, you may want to divide the contents into smaller aliquots, such as 10μ L per tube, and keep them at -20°C. Avoid repeatedly freezing and thawing the product.
- Using this product is easy and convenient since you don't need to heat it up first. However, be aware of the electrophoretic buffer and gel preparation to prevent any issues with the results. You can safely store the product at 4°C for 3 months, but if you need to keep it for longer, store it at -20°C. To avoid contamination, divide the product into smaller tubes and freeze them at -20°C. Each tube can contain 10μ L, which is a suitable amount for each use. Don't freeze and thaw the product repeatedly.
- No heating is required for using this product. To ensure the electrophoretic results are accurate, change the buffer regularly and use a freshly prepared gel. For storage, the product can be kept at 4°C for up to 3 months or at -20°C for longer periods. To avoid contamination, divide the product into smaller tubes containing 10μ L each, and freeze them at -20°C. Use one tube at a time to avoid thawing and refreezing.
Quality Control
Under suggested conditions, Prestained Protein Ladder resolves 12 major bands in 15% SDS-PAGE (Trisglycine buffer) and after Western blotting to nitrocellulose membrane.
