Description

Description
Immunohistochemical staining involves the use of endogenous alkaline phosphatase blocking solution, also known as inactivation solution, which primarily consists of levamisole/thiamidazole hydrochloride. This solution can be mixed with a developer for the staining process. Alkaline phosphatase is commonly employed in ELISA detection, producing a soluble colored substance as its reaction product. This colored substance is often used for quantitative determination using a spectrophotometer. However, the remaining activity of alkaline phosphatase, although reduced by paraffin embedding, can still impact the staining results. Our company offers an alkaline phosphatase blocking solution (Levamisole method) containing levamisole, preservatives, and other components. This solution effectively eliminates alkaline phosphatase interference in the developer solution, except for small intestinal isomers.
Usage
After being attached to the glass slide, the section undergoes a gentle wash with PBS. It is crucial to follow this procedure in order to ensure the accuracy and quality of the sample. The washing process helps to remove any unwanted particles or debris that could interfere with the analysis and interpretation of the sample. Therefore, it is important to handle the section with care and accuracy during this critical stage of the experiment.
To prepare the slides for further processing, it is important to blot or blow dry them first. Then, they should be placed in an alkaline phosphatase blocking solution using the Levamisole method. This process involves incubating the slides for a period of around 10 to 20 minutes to allow complete blocking of the enzyme. By following these steps, the slides will be ready for subsequent analysis or experimentation.
Sure! Instead of cleaning, you can simply use a vacuum or hairdryer to remove any debris from the slide.
4. Incubate in unlabeled primary antibody.
General Notes
To ensure your optimal health and safety, we kindly request that you put on disposable gloves and lab coats.
