Endogenous Biotin Blocking Solution

When tissues are fixed with formalin, the naturally occurring background from endogenous biotin is reduced. However, in frozen sections, the background is significantly increased. Aggressive antigen retrieval methods can expose endogenous substances in formalin-fixed tissues, such as endogenous avidin-biotin. To counteract this, a reagent containing an endogenous biotin blocking solution is used. This solution effectively inhibits the activity of endogenous substances, primarily endogenous biotin. It is particularly useful for blocking the interference caused by endogenous substances in immunohistochemistry. 1. Dewaxing and hydration of sections
Afterwards, proceed to add the endogenous biotin blocking solution to the sample and allow it to incubate for a duration of 20 minutes.
3. Wash in PBS or Tris buffer for 5 minutes.
4. Perform immunostaining steps.

It is advisable to opt for a detection system that does not involve streptavidin-biotin, in case the tissue sample being tested possesses abundant endogenous streptavidin-biotin content.
