| Description | Antigen retrieval is a crucial process in tissue production as it allows the exposure of antigens that may have been blocked by chemical reagents or distorted due to heat. This process involves using buffers like citrate, EDTA, or Tris to re-expose the antigens without damaging the epitopes. In the past, trypsin, proteinase K, and pepsin were used to induce epitope retrieval. Nowadays, the company's pepsin antigen retrieval solution has greatly improved the immunostaining effect of paraffin sections. Antigen retrieval is particularly important for paraffin sections while frozen sections may not necessarily require it. However, antigen retrieval can still enhance the staining effect of frozen sections in some cases. To retrieve antigens for 10 samples, 10ml antigen retrieval solution (1×) is required for each film with a total of 100ml antigen retrieval solution. With antigen retrieval, the detection rate of antigens can increase while background staining and diagnosis accuracy can improve significantly. |
| Usage | Operation steps (for reference only): 1. Paraffin section 1. Dewax to water (1) Xylene 3 times, 3~5min each time. To perform dehydration with absolute ethanol, repeat the process twice for 3 to 5 minutes each time. This will ensure proper dehydration of the material. The use of absolute ethanol is necessary to effectively remove any remaining moisture, and it is important to perform this step as directed for optimal results. (3) 95% ethanol, 3~5min. (4) 90% ethanol, 3~5min. (5) 80% ethanol, 3~5min. (6) 70% ethanol, 3~5min. Please follow the procedure and rinse the material twice with distilled water. Each rinse should last approximately 3 to 5 minutes. To retrieve the antigens, you need to immerse the slices in a pepsin antigen retrieval solution. This solution should be incubated at 37°C for a time ranging from 10-25 minutes. This process will help prepare the tissue for further examination. It is important to follow these steps carefully to ensure accurate results. Antigen retrieval is a critical step in detecting specific proteins in tissues and is commonly used in immunohistochemistry. 3. Wash with immunostaining detergent or tap water for 1 to 2 times, 3 to 5 minutes each time, and rinse thoroughly. 4. Perform subsequent immunostaining steps. 2. Frozen section: 1. Wash the sections with immunostaining washing solution or tap water for 5 minutes. 2. Antigen retrieval: Immerse the slices in pepsin antigen retrieval solution and incubate at 37°C for 10-25 minutes. 3. Wash with immunostaining detergent or tap water for 1 to 2 times, 3 to 5 minutes each time, and rinse thoroughly. 4. Perform subsequent immunostaining steps 3.
