Testing Principle:

The double antibody sandwich ELISA technique is employed in this kit. The conjugate plate is preloaded with a specific anti-human CXCL13 Capture Antibody, which exhibits high affinity. To begin the assay, the standard substance, the sample being tested, and the biotin-labeled detection antibody are added sequentially into the HRP-plate well. The mixture is thoroughly oscillated and mixed, allowing for a 2-hour incubation period at room temperature. During this incubation step, the CXCL13 in the sample combines with the solid phase antibody and the detection antibody. After incubation, the plate is thoroughly washed to eliminate any unbound or free components. Next, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added to the plate. Following another round of washing, a TMB color substrate is introduced and allowed to incubate in the dark at room temperature for color development. The depth of the color reaction is directly correlated to the concentration of CXCL13 present in the sample. To terminate the reaction, a termination solution is added, and the absorbance value is measured at a detection wavelength of 450 nm using a microplate analyzer. It is important to note that during measurement, a correction wavelength of 570-630 nm is utilized.
