Testing Principle：

The double antibody sandwich ELISA is utilized in this kit, utilizing a specific anti-human CXCL13 Capture Antibody that comes prepackaged on a high affinity conjugate plate. To perform the test, the standard substance, the sample that needs to be tested, and the biotin labeled detection antibody are consecutively added to the HRP-plate well. After proper mixing and oscillation, the CXCL13 in the sample goes through a 2-hour incubation process at room temperature, which allows it to combine with the solid phase antibody and detection antibody.
Next, the mixture is thoroughly washed to remove any unbound ingredients. Subsequently, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added, and after another round of washing, TMB color substrate is added. The mixture is then incubated in the dark at room temperature to allow color development, where the depth of the color reaction is linked to the concentration of CXCL13 in the sample.
To terminate the reaction, a termination solution is added, and the absorbance value is measured at 450 nm detection wavelength using a microplate analyzer, with correction wavelength 570-630 nm. By using this method, researchers can accurately test for the presence of CXCL13.

Background：

IL-11 is a cytokine that was initially isolated from bone marrow mesenchymal stem cells nearly three decades ago. It belongs to the IL-6 cytokine family and is characterized by the use of the co-receptor gp130. Its primary role is in hematopoietic regulation and specifically in the maturation of megakaryocytes. Moreover, it has been shown to be effective in mitigating chemotherapy-induced laminopenia, regulating antigen-antibody response, and playing a crucial role in bone cell proliferation and differentiation. IL-11 functions as a signaling molecule and has various effects, including placental formation and partly decidualization, playing a key role in blastocyst implantation into the endometrium. It stimulates the growth of specific lymphocytes and causes bone resorption, while in a mouse model, it increases the thickness and strength of the long bone cortex. Oprexil interleukin, a recombinant form of IL-11, is widely used in cancer patients to prevent severe thrombocytopenia.
