The Caspase 6 activity detection kit utilizes spectrophotometry to identify caspase 6 enzymatic activity in cell or tissue lysate, as well as purified caspase 6 enzymatic activity. Caspase enzymes are a vital group of proteases involved in cellular apoptosis. Caspase 6 can be activated by granzyme B through cleavage of its precursor, leading to the formation of an active caspase 6 dimer. It has been observed that activated caspase 6 plays a crucial role in inducing cell apoptosis. Additionally, caspase 6 has the ability to cleave specific proteins including PARP, keratin-18, and Lamin A, an important component of the nuclear envelope. Notably, Lamin A can only be cleaved by caspase 6 within the caspase family. The recognition site on Lamin A for caspase 6 is VEID. In this detection kit, the specificity of caspase 6 for the VEID recognition site is utilized, and a caspase 6-specific chromogenic substrate called Ac-VEID-pNA is designed accordingly.
