Data Examples

For the usage of IP antibody, we recommend using 5ug of His Mouse IgG1 per ml Lysate for Western blotting at a dilution of 1:3000. We suggest using 5 different samples for your experiments, which include untransfected 293 cell lysate, transfected 293 cell lysate with His-tag fusion protein, and 3 different types of immune precipitation. The first immune precipitation will include untransfected 293 cells with anti-His monoclonal antibody and Protein G agarose, the second will have transfected 293 cells with normal Mouse IgG and Protein G agarose, and the third will contain transfected 293 cells with anti-His monoclonal antibody and Protein G agarose. Lastly, we recommend using recombinant protein (E.coli) sample for comparison in your experiments. By including these different samples, you can determine if the antibody accurately detects the His-tag fusion protein in the transfected cell lysate and also examine potential non-specific binding in different controls.
