Description

The active component, Upf1, plays a critical role in the mRNA surveillance mechanism, nonsense-mediated decay (NMD), responsible for degrading mRNAs containing premature termination codons. Identified as an ATP-dependent RNA helicase in the cytoplasm, Upf1 later discovered to be a component of cytoplasmic P-bodies. The Upf1 phosphorylation process allows for the repression of translation and mRNA accessibility to the NMD machinery during NMD. Two other crucial components of NMD, Upf2 and Upf3, were also identified and found to have distinct localizations, with Upf2 located perinuclearly and Upf3 located nucleocytoplasmically.

Other namesRENT1 is an essential ATP-dependent helicase that plays a crucial role in regulating nonsense-mediated mRNA decay (NMD). It is also known as Delta helicase, FLJ43809, FLJ46894, HUPF 1, kIAA0221, or Smg 2 homolog NMD factor. RENT1 is responsible for recognizing premature termination codons (PTCs) in mRNA, triggering NMD to eliminate these abnormal transcripts to maintain genome integrity and prevent the formation of truncated or defective proteins.
In addition to its role in NMD, RENT1 also participates in other cellular processes, including RNA splicing, translation initiation, and ribosome biogenesis. It has been found to interact with numerous proteins, including UPF2, UPF3, SMG1, eIF4B, and RNPS1, to regulate various aspects of RNA metabolism.
RENT1 is highly conserved across species, with homologs found in yeast (Upf1p) and other eukaryotes. Mutations in RENT1 have been associated with several human diseases, including cancer, neurodegeneration, and intellectual disability. Therefore, understanding the role and regulation of RENT1 is critical for developing novel therapeutic approaches to address these diseases.

SpecificityThe total UPF1 levels in an organism can be effectively detected by the UPF1 Antibody. The antibody is designed to recognize and bind to the endogenous UPF1 proteins present in the biological system. This detection method allows researchers to study the overall expression of UPF1 and its role in various cellular processes. By utilizing the UPF1 Antibody, scientists can gain valuable insights into the abundance and regulation of UPF1 in their experimental models.

ApplicationFor Western blot, the recommended antibody dilution range is 1:500-1:2000. When performing immunohistochemistry, a dilution range of 1:50-1:200 is suggested. For immunofluorescence or immunocytochemistry, dilute the antibody at 1:100-1:500. For ELISA with peptide antigen, the recommended dilution range is 1:20000-1:40000. It is important to follow these recommended dilution ranges to ensure optimal results.

Target

Background

RNA-dependent helicase and ATPase required for nonsense-mediated decay (NMD) of mRNAs containing premature stop codons. Is recruited to mRNAs upon translation termination and undergoes a cycle of phosphorylation and dephosphorylation; its phosphorylation appears to be a key step in NMD. Recruited by release factors to stalled ribosomes together with the SMG1C protein kinase complex to form the transient SURF (SMG1-UPF1-eRF1-eRF3) complex. In EJC-dependent NMD, the SURF complex associates with the exon junction complex (EJC) (located 50-55 or more nucleotides downstream from the termination codon) through UPF2 and allows the formation of an UPF1-UPF2-UPF3 surveillance complex which is believed to activate NMD. Phosphorylated UPF1 is recognized by EST1B/SMG5, SMG6 and SMG7 which are thought to provide a link to the mRNA degradation machinery involving exonucleolytic and endonucleolytic pathways, and to serve as adapters to protein phosphatase 2A (PP2A), thereby triggering UPF1 dephosphorylation and allowing the recycling of NMD factors. UPF1 can also activate NMD without UPF2 or UPF3, and in the absence of the NMD-enhancing downstream EJC indicative for alternative NMD pathways. Plays a role in replication-dependent histone mRNA degradation at the end of phase S; the function is independent of UPF2. For the recognition of premature termination codons (PTC) and initiation of NMD a competitive interaction between UPF1 and PABPC1 with the ribosome-bound release factors is proposed. The ATPase activity of UPF1 is required for disassembly of mRNPs undergoing NMD. Essential for embryonic viability.
