Description

Troponin is a crucial component that, together with tropomyosin, regulates muscle contraction in response to changes in Ca2+ levels within the cell. It consists of three subunits: troponin C (TnC), which binds to Ca2+; troponin T (TnT), which binds to tropomyosin; and troponin I (TnI), which inhibits muscle contraction. When the heart is stimulated by β-adrenergic signals, TnI (cardiac) is phosphorylated by PKA and PKC at Ser23 and Ser24. This phosphorylation triggers a change in the shape of TnC's regulatory domain, leading to decreased interaction between TnI and TnC. Consequently, the binding affinity of TnC for Ca2+ decreases, resulting in reduced sensitivity of myofilaments to Ca2+ (1-3).
Different isoforms of TnT exist in various muscle types: slow skeletal muscle (ssTnT/TNNT1), fast skeletal muscle (fsTnT/TNNT3), and cardiac muscle (cTnT/TNNT2), each potentially having multiple alternatively spliced variants. Detecting the serum levels of cTnT and cTnI through assays has proven valuable in analyzing cardiac injuries.

Troponin T is the tropomyosin-binding subunit of troponin, the thin filament regulatory complex which confers calcium-sensitivity to striated muscle actomyosin ATPase activity.

Tissue specificityHeart. The fetal heart shows a greater expression in the atrium than in the ventricle, while the adult heart shows a greater expression in the ventricle than in the atrium. Isoform 6 predominates in normal adult heart. Isoforms 1, 7 and 8 are expressed in fetal heart. Isoform 7 is also expressed in failing adult heart.
