Description | Troponin is a crucial component that, together with tropomyosin, regulates muscle contraction in response to changes in Ca2+ levels within the cell. It consists of three subunits: troponin C (TnC), which binds to Ca2+; troponin T (TnT), which binds to tropomyosin; and troponin I (TnI), which inhibits muscle contraction. When the heart is stimulated by β-adrenergic signals, TnI (cardiac) is phosphorylated by PKA and PKC at Ser23 and Ser24. This phosphorylation triggers a change in the shape of TnC's regulatory domain, leading to decreased interaction between TnI and TnC. Consequently, the binding affinity of TnC for Ca2+ decreases, resulting in reduced sensitivity of myofilaments to Ca2+ (1-3). |
