Description | The Toll-like receptor (TLR) family plays a crucial role in innate immune responses by recognizing various pathogens and triggering defense responses. These receptors are named after their close relative Toll in Drosophila. Upon activation, TLRs associate with cytoplasmic adaptor proteins containing TIR domains, such as MyD88, MAL/TIRAP, TRIF, and TRAM, which lead to the recruitment and activation of IRAK1 and IRAK4. This complex then activates TAK1 and IKK, leading to the degradation of IκB that normally maintains NF-κB inactivity in the cytoplasm. TLR7, 8, and 9 are a group of structurally related TLRs localized to intracellular endosomes, with TLR7 showing the highest expression in lung, placenta, and spleen. TLR7 mediates responses to both synthetic compounds, including imidazoquinolines, and ssRNA viruses, which activate NF-κB and trigger IFN production. The TLRs and members of the IL-1 receptor family share a conserved stretch of approximately 200 amino acids known as the TIR domain. |
