Description

Type I, II and IV collagens are all bound by TGFBI, a protein containing the RGD motif. The RGD motif is found in many extracellular matrix proteins, which aid in cell adhesion, and it is also a ligand recognition sequence for several integrins. It is believed that TGFBI plays a role in the interactions between cells and collagen, and it may even be involved in endochondrial bone formation within cartilage. Transforming growth factor-beta induces the production of TGFBI, and its main function is to inhibit cell adhesion.

Other namesThe protein known as RGD containing collagen associated protein goes by many names, including AI181842, AI747162, Beta ig, Beta ig h3, and BGH3_HUMAN. It is also sometimes referred to as Big h3, BIGH3, CDB1, CDG2, CDGG1, CSD, CSD1, CSD2, CSD3, EBMD, Kerato epithelin, Kerato-epithelin, LCD1, MGC150270, RGD CAP, RGD-CAP, and RGD-containing collagen-associated protein. This versatile protein is also known as TGFBI transforming growth factor, beta induced, 68kDa, and Transforming growth factor beta induced protein ig h3, as well as Transforming growth factor-beta-induced protein ig-h3. Despite all these different names, the underlying function of RGD containing collagen associated protein remains the same.
SourceRabbit
SpecificityThe TGFBI Antibody is capable of detecting the total amount of TGFBI present in a biological sample, without the need for artificial changes or manipulations. This allows for the accurate analysis of endogenous levels of TGFBI, producing highly reliable results.
Species ReactivityHuman;Mouse;Rat
Predictive reaction speciesPig;Rabbit;Sheep;Dog;Bovine
AntigenTGFBI
ApplicationThe dilution ratios for Western blot (WB) range from 1:500 to 1:2000, while for Immunohistochemistry (IHC) they range from 1:50 to 1:200. For ELISA (peptide), the dilution range is between 1:20000 and 1:40000. These dilution ratios provide guidelines for the appropriate concentrations of antibodies or peptides to use in each assay. It is essential to follow these dilution ranges to ensure accurate and reliable results in protein analysis.
ImmunogenA synthetic peptide of human TGFBI.

Target

Background

Plays a role in cell adhesion (PubMed:8024701). May play a role in cell-collagen interactions (By similarity).

Tissue specificityHighly expressed in the corneal epithelium (PubMed:27609313, PubMed:8077289). Expressed in heart, placenta, lung, liver, skeletal muscle, kidney and pancreas (PubMed:8077289).
Posttranslational modificationGamma-carboxylation is controversial. Gamma-carboxyglutamated; gamma-carboxyglutamate residues are formed by vitamin K dependent carboxylation; these residues may be required for binding to calcium (PubMed:18450759). According to a more recent report, does not contain vitamin K-dependent gamma-carboxyglutamate residues (PubMed:26273833).The EMI domain contains 2 expected intradomain disulfide bridges (Cys-49-Cys85 and Cys-84-Cys-97) and one unusual interdomain disulfide bridge to the second FAS1 domain (Cys-74-Cys-339). This arrangement violates the predicted disulfide bridge pattern of an EMI domain.
Celluar localizationExtracellular region or secreted;Golgi apparatus;Plasma Membrane;
