Description | This gene is responsible for producing a protease inhibitor that plays a crucial role in regulating the tissue factor (TF)-dependent pathway involved in blood coagulation. Blood coagulation begins with the formation of a complex between factor VIIa and TF, which then activates other proteases (factors IX and X) in a sequential manner, ultimately leading to the creation of a fibrin clot. The protein produced by this gene acts as an inhibitor for the activated factor X and VIIa-TF proteases, establishing a self-regulatory mechanism. It is worth noting that the protein is glycosylated and primarily found in the vascular endothelium and plasma, existing both in its free form and bound to plasma lipoproteins. Multiple variations of this gene, resulting from alternative splicing of its transcripts, have been identified. However, some of these variants have not yet been fully characterized. It is also referred to as anti-convertin, EPI, extrinsic pathway inhibitor, LACI and TFI. The human TFPI is encoded by the TFPI1 gene. This protein plays a crucial role in regulating the extrinsic pathway of coagulation. Mutations in this gene cause an increased risk of thrombosis. TFPI1 is a powerful anticoagulant and its deficiency is associated with a predisposition to clotting disorders. In summary, TFPI is a vitally important protein involved in preventing blood clotting disorders and maintaining the proper function of the body's circulatory system. |
