Description | Epithelial cell adhesion and activating molecule (EpCAM/CD326) is a transmembrane glycoprotein responsible for mediating Ca2+-independent, homophilic adhesions on the basolateral surface of most epithelial cells. Although EpCAM is not expressed in adult squamous epithelium, it is highly expressed in adeno and squamous cell carcinomas, making it a valuable marker of epithelial and tumor tissue. In fact, EpCAM was one of the first tumor-associated antigens identified through research studies, and it has since been shown to be highly expressed in cancer cells, as evidenced by numerous studies and reviews. This indicates the potential clinical utility of EpCAM as a diagnostic and therapeutic target for cancer treatment. |
| Other names | The Epithelial Cell Adhesion Molecule, also known as EpCAM or CD326, is a glycoprotein located on the surface of epithelial cells. It has numerous alternative names, including Adenocarcinoma-associated antigen and Tumor-associated calcium signal transducer 1. EpCAM is involved in cell adhesion, migration, proliferation, and differentiation. It is expressed in various epithelia, such as the gastrointestinal tract, liver, and breast, and is also present in circulating tumor cells. Studies have shown that abnormal expression of EpCAM is associated with cancer development and progression, making it a target for cancer therapy. EpCAM is therefore a valuable biomarker for cancer diagnosis and treatment. |

Target | |
Background | May act as a physical homophilic interaction molecule between intestinal epithelial cells (IECs) and intraepithelial lymphocytes (IELs) at the mucosal epithelium for providing immunological barrier as a first line of defense against mucosal infection. Plays a role in embryonic stem cells proliferation and differentiation. Up-regulates the expression of FABP5, MYC and cyclins A and E. |
| Tissue specificity | Highly and selectively expressed by undifferentiated rather than differentiated embryonic stem cells (ESC). Levels rapidly diminish as soon as ESC's differentiate (at protein levels). Expressed in almost all epithelial cell membranes but not on mesodermal or neural cell membranes. Found on the surface of adenocarcinoma. |
| Posttranslational modification | Hyperglycosylated in carcinoma tissue as compared with autologous normal epithelia. Glycosylation at Asn-198 is crucial for protein stability. |
| Celluar localization | Extracellular region or secreted;Plasma Membrane; |
| UniPort | P16422 |
