Overview | |
Description | The process of transcribing protein-coding genes can be carried out effectively on naked DNA with only the involvement of general transcription factors and RNA polymerase II. However, when DNA is packed into chromatin, transcription cannot occur without the aid of accessory factors. One such crucial factor is called FACT, which stands for "facilitates chromatin transcription." FACT has the ability to interact specifically with histones H2A/H2B, which helps to disassemble nucleosomes and allows for smooth transcription elongation. The two main subunits of FACT are the 80 kDa and 140 kDa proteins. The latter subunit is the focal point of the gene that encodes for FACT. Through its unique functions, FACT plays an essential role in promoting proper gene transcription on chromatinized DNA. |
Other names | Chromatin-specific transcription elongation factor 140 kDa subunit, also known as CDC68, is an important component of the chromatin transcription complex. This 140 kDa subunit, together with the chromatin-specific transcription elongation factor, plays a crucial role in facilitating transcriptional processes. It is also referred to as the FACT 140 kDa subunit or FACTp140. |

Stability & Storage | To preserve the product's integrity, it is recommended to store it at a temperature of -20 °C for up to one year. It is important to avoid subjecting the product to repeated cycles of freezing and thawing, as this can compromise its quality. |
Storage buffer | The recommended storage condition for Rabbit IgG is at -20 °C in phosphate buffered saline with a pH of 7.4. The solution should contain 150mM NaCl, 0.02% sodium azide, and 50% glycerol. It is advised to store the product at this temperature to maintain its stability for up to 12 months from the date of receipt. |

Background | The FACT complex, a vital component, plays a crucial role in chromatin reorganization by acting as a general factor. It is actively involved in numerous DNA-dependent processes such as DNA replication, mRNA elongation, and DNA repair. Specifically during transcription elongation, the FACT complex acts as a histone chaperone, effectively disrupting and then restoring the structure of nucleosomes. This enables the smooth passage of RNA polymerase II and facilitates transcription by promoting the dissociation of a histone H2A-H2B dimer from the nucleosome. Furthermore, it subsequently aids in the reestablishment of the nucleosome after RNA polymerase II has passed through. There is a possibility that the FACT complex is also associated with CK2 (casein kinase II), leading to the phosphorylation of 'Ser-392' residue on p53/TP53. |
Tissue specificity | Ubiquitous. |
Posttranslational modification | ADP-ribosylated. ADP-ribosylation by PARP1 is induced by genotoxic stress and correlates with dissociation of FACT from chromatin. |
Celluar localization | Nucleus; |
