This important protein is encoded by the POLR1B gene and plays a crucial role in transcribing ribosomal RNA (rRNA) in the nucleoli of eukaryotic cells. The function of polymerase (RNA) I polypeptide B 128kDa is to catalyze the synthesis of a large precursor RNA molecule known as pre-rRNA. The pre-rRNA is then processed to yield the mature 18S, 5.8S, and 28S rRNAs, which form the basic structure of the ribosome. Therefore, RNA polymerase I polypeptide B is essential for the correct function of ribosomes and, by extension, protein synthesis in all eukaryotic organisms. The DNA-directed RNA polymerase I subunit rpa2 is a large protein that is composed of over 1,100 amino acids. It is an essential component of the RNA polymerase I complex and interacts with several other subunits to form a stable polymerase machinery. Various mutations in the POLR1B gene have been associated with several human diseases, including Treacher Collins syndrome, an autosomal dominant craniofacial disorder.
