Description

The GTPase-activating protein (GAP) family encompasses the protein encoded by this specific gene. These GAPs have a remarkable ability to bind to activated forms of Rho GTPases and promote the hydrolysis of GTP. By virtue of this catalytic function, GAPs exert a negative regulatory effect on Rho-mediated signals. Multiple essential roles are attributed to this protein, including its participation in the control of cytokinesis initiation, regulation of cell growth and differentiation of hematopoietic cells, modulation of spermatogenesis, and facilitation of neuronal proliferation. Interestingly, various alternatively spliced transcript variants have been identified for this gene, adding another layer of complexity to its functional diversity.

Other namesCYK4, also known as Rac GTPase-activating protein 1 or RGAP1, is a protein that plays a crucial role in regulating cell division and cytokinesis. It is encoded by the KIAA1478 gene and is expressed in multiple tissues including male germ cells. CYK4 functions as a GTPase activating protein (GAP), which helps in the hydrolysis of GTP to GDP and thereby regulates the activity of small GTPases like Rac1. At the cellular level, CYK4 interacts with several other proteins and forms a complex called the centralspindlin complex, which is essential for proper spindle assembly and cytokinesis. Dysregulation of CYK4 expression or function has been implicated in various diseases including cancer. Therefore, CYK4 represents a potential therapeutic target for the treatment of these diseases.

SpecificityThe RACGAP1 Antibody is capable of identifying the complete quantity of RACGAP1 that exists naturally within an organism. This antibody detects endogenous levels of total RACGAP1 and serves as a reliable tool for conducting research in the field. By rearranging the original content, we can convey the same information in a slightly different manner.

AntigenRACGAP1
ApplicationFor western blotting, a dilution of 1:500 to 1:2000 is recommended, while for immunohistochemistry the range is 1:50 to 1:200. If you're using fluorescence-based techniques like immunofluorescence or immunocytochemistry, the suggested dilution is 1:100 to 1:500. If you need to perform an ELISA with a peptide substrate, dilute your antibody to between 1:20000 and 1:40000. It's important to follow the recommended dilutions to ensure accurate and reliable results.

Target

Background

Component of the centralspindlin complex that serves as a microtubule-dependent and Rho-mediated signaling required for the myosin contractile ring formation during the cell cycle cytokinesis. Required for proper attachment of the midbody to the cell membrane during cytokinesis. Plays key roles in controlling cell growth and differentiation of hematopoietic cells through mechanisms other than regulating Rac GTPase activity. Also involved in the regulation of growth-related processes in adipocytes and myoblasts. May be involved in regulating spermatogenesis and in the RACGAP1 pathway in neuronal proliferation. Shows strong GAP (GTPase activation) activity towards CDC42 and RAC1 and less towards RHOA. Essential for the early stages of embryogenesis. May play a role in regulating cortical activity through RHOA during cytokinesis. May participate in the regulation of sulfate transport in male germ cells.

Tissue specificityHighly expressed in testis, thymus and placenta. Expressed at lower levels in spleen and peripheral blood lymphocytes. In testis, expression is restricted to germ cells with the highest levels of expression found in spermatocytes. Expression is regulated in a cell cycle-dependent manner and peaks during G2/M phase.
Posttranslational modificationPhosphorylated at multiple sites in the midbody during cytokinesis. Phosphorylation by AURKB on Ser-387 at the midbody is, at least in part, responsible for exerting its latent GAP activity towards RhoA. Phosphorylation on multiple serine residues by PLK1 enhances its association with ECT2 and is critical for cleavage furrow formation.
