Description | S5a (PSMD4) is a pivotal component of the 19S regulatory proteasome complex, which plays a crucial role in the recognition and degradation of ubiquitinated proteins (1). With its two polyubiquitin binding motifs (UIM), S5a can selectively bind to multiubiquitin chains through hydrophobic interaction (2,3). Interestingly, the UIM of S5a exhibits a remarkable affinity not only for ubiquitin but also for a ubiquitin-like domain found in various proteins. By directly interacting with these proteins, S5a serves as a crucial mediator in directing them towards the proteasome for subsequent degradation (4,5). |
| Other names | Proteasome subunit S5a, also known as 26S proteasome regulatory subunit rpn10 or multisubstrate protein ASF, is a non-ATPase regulatory subunit of the 26S proteasome. It has been implicated in several cellular processes, including protein degradation, cell division, and transcriptional regulation. S5a is a multiubiquitin chain-binding protein that recognizes and binds to ubiquitinated proteins, targeting them for proteasomal degradation. This process plays a critical role in maintaining cellular homeostasis by regulating the turnover of damaged or misfolded proteins. In addition to its role in protein degradation, S5a has also been shown to interact with and regulate the activity of several transcription factors, including P53 and NF-κB. These interactions suggest that S5a may play a broader role in the regulation of gene expression. Overall, S5a is a versatile protein that plays a critical role in maintaining cellular homeostasis and regulating key cellular processes. Further research on the functions of this protein will undoubtedly reveal even more insights into its importance in cellular biology. |

Background | Component of the 26S proteasome, a multiprotein complex involved in the ATP-dependent degradation of ubiquitinated proteins. This complex plays a key role in the maintenance of protein homeostasis by removing misfolded or damaged proteins, which could impair cellular functions, and by removing proteins whose functions are no longer required. Therefore, the proteasome participates in numerous cellular processes, including cell cycle progression, apoptosis, or DNA damage repair. PSMD4 acts as an ubiquitin receptor subunit through ubiquitin-interacting motifs and selects ubiquitin-conjugates for destruction. Displays a preferred selectivity for longer polyubiquitin chains. |
