Description

PIK3R1 functions as a regulatory subunit of phosphoinositide-3-kinase and is responsible for mediating binding to specific tyrosine-phosphorylated proteins via its SH2 domain. It plays a critical adapter role in the association of alpha, beta, and delta enzymes' p110 catalytic units to the plasma membrane, facilitating the phosphorylation of inositol lipids by p110. Additionally, it may play a role in regulating the actin cytoskeleton.
In insulin-sensitive tissues, PIK3R1 is essential for the insulin-induced increase in glucose uptake and glycogen synthesis. Essentially, the protein serves as a sort of mediator, allowing the catalytic unit of key enzymes to interact with the appropriate molecules and carry out their crucial functions. By doing so, PIK3R1 helps to maintain proper cellular signaling and metabolic activity, ultimately contributing to overall health and well-being.

Other namesGRB1, also known as p85 alpha, is a regulatory subunit of phosphatidylinositol 3 kinase (PI3K). It is involved in cellular signaling pathways and plays a crucial role in regulating cell growth, proliferation, and survival. The full name of this protein is Phosphatidylinositol 3 kinase associated p85 alpha, and it is encoded by the gene PIK3R1.
On the other hand, there is another regulatory subunit, known as p55 gamma or p85 gamma, which is encoded by the gene PIK3R3. This subunit, also a part of PI3K, is called Phosphatidylinositol 3 kinase regulatory subunit gamma. It is involved in similar cellular signaling pathways as p85 alpha.
Both p85 alpha and p85 gamma regulate the activity of the catalytic subunit of PI3K, enabling the conversion of phosphatidylinositol 4,5-bisphosphate (PIP2) to phosphatidylinositol 3,4,5-trisphosphate (PIP3). PIP3 acts as a secondary messenger and activates downstream signaling cascades that regulate various cellular processes such as cell growth, differentiation, and survival.
In summary, GRB1 or p85 alpha, and p55 gamma or p85 gamma are regulatory subunits of PI3K. They play key roles in modulating cellular signaling pathways and are essential for maintaining proper cell functions.

Target

Background

Binds to activated (phosphorylated) protein-Tyr kinases, through its SH2 domain, and acts as an adapter, mediating the association of the p110 catalytic unit to the plasma membrane. Necessary for the insulin-stimulated increase in glucose uptake and glycogen synthesis in insulin-sensitive tissues. Plays an important role in signaling in response to FGFR1, FGFR2, FGFR3, FGFR4, KITLG/SCF, KIT, PDGFRA and PDGFRB. Likewise, plays a role in ITGB2 signaling (PubMed:17626883, PubMed:19805105, PubMed:7518429). Modulates the cellular response to ER stress by promoting nuclear translocation of XBP1 isoform 2 in a ER stress- and/or insulin-dependent manner during metabolic overloading in the liver and hence plays a role in glucose tolerance improvement (PubMed:20348923).

Tissue specificityIsoform 2 is expressed in skeletal muscle and brain, and at lower levels in kidney and cardiac muscle. Isoform 2 and isoform 4 are present in skeletal muscle (at protein level).
Posttranslational modificationPolyubiquitinated in T-cells by CBLB; which does not promote proteasomal degradation but impairs association with CD28 and CD3Z upon T-cell activation.Phosphorylated. Tyrosine phosphorylated in response to signaling by FGFR1, FGFR2, FGFR3 and FGFR4. Phosphorylated by CSF1R. Phosphorylated by ERBB4. Phosphorylated on tyrosine residues by TEK/TIE2. Dephosphorylated by PTPRJ. Phosphorylated by PIK3CA at Ser-608; phosphorylation is stimulated by insulin and PDGF. The relevance of phosphorylation by PIK3CA is however unclear (By similarity). Phosphorylated in response to KIT and KITLG/SCF. Phosphorylated by FGR.
