Overview

Description

Mnk1, a member of the MAPKKAPK family of CAM kinases, plays a role in regulating translation. Its activation by MAP kinases leads to the phosphorylation of eIF4E at Ser209, which in turn enhances its binding affinity for mRNA caps containing 7-methylguanosine. The kinase activity of mouse Mnk1 relies on two crucial phosphorylation sites, Thr197 and Thr202. Additionally, three different isoforms of Mnk1 have been identified in humans through alternative splicing.

Other names

Interacting with MAP kinases, MAP kinase interacting kinase 1 (MKNK1) also known as MAP kinase interacting serine/threonine kinase 1 and MAP kinase signal integrating kinase 1 functions as a signal-integrating kinase. MKNK1 plays a crucial role in mediating the cellular response to growth factors and stress signals. It interacts with MAP kinases to regulate various cellular processes. Additionally, MKNK1 is involved in phosphorylation of serine/threonine residues in target proteins, thereby modulating their activity. Known as MNK1 or MITOGEN-ACTIVATED PROTEIN KINASE-INTERACTING SERINE/THREONINE KINASE 1, this protein is encoded by the MKNK1 gene in humans. Understanding the functions and regulatory mechanisms of MKNK1 is vital for elucidating its role in cell signaling and its potential implications in disease pathogenesis.

Source

Rabbit

Specificity

The MKNK1 antibody is capable of detecting total levels of MKNK1 that are present naturally within the body. This means that it is able to recognize the protein regardless of its source, whether it be from cells that are in their natural state or ones that have been otherwise altered. By utilizing this antibody, researchers can gain valuable insights into the behavior and function of MKNK1 in a variety of biological contexts.

Stability & Storage

To ensure optimal storage conditions, it is recommended to keep the product at a temperature of -20 °C for up to one year. It is important to avoid subjecting the product to repeated freeze/thaw cycles. By adhering to these storage guidelines, the product will maintain its integrity and remain viable for future use.

Storage buffer

This rabbit IgG is suspended in a solution of phosphate buffered saline with a pH of 7.4. It contains 150mM NaCl and 0.02% sodium azide, along with 50% glycerol. To maintain its stability, it should be stored at a temperature of -20 °C. This product has a shelf life of 12 months from the date of receipt.

Target

Background

This protein may have a key role in responding to environmental stress and cytokines. It is believed to regulate translation by phosphorylating EIF4E, which consequently increases the protein's affinity for mRNA cap containing 7-methylguanosine. This mechanism potentially provides a subtle yet important control of the translation process in response to various environmental stresses.

Tissue specificity

Ubiquitous.

Posttranslational modification

To activate the kinase, dual phosphorylation of Thr-250 and Thr-255 is necessary, as well as phosphorylation of Thr-385. One of the kinases that triggers the activation of MKNK1/MNK1 is MAPK3/ERK1. Conversely, PAK2-mediated phosphorylation results in decreased EIF4G1 phosphorylation. It is important to note that these modifications all play a critical role in regulating the activity of the kinase.
