Description

Midkine, also known as MK, plays a crucial role in regulating growth and differentiation during embryonic development. It is a heparin-binding molecule with a mature human protein consisting of 118 amino acids. MK is a non-glycosylated protein that shares more than 87% identity between human and mouse.
In embryonic development, MK expression is closely regulated by steroid receptors of the retinoic acid superfamily. Its carboxy-terminus contains the primary heparin-binding site and the neurite-promoting sequences, responsible for its neurotrophic properties. Both the amino- and carboxy-terminal sequences are required for these properties.
Interestingly, MK overexpression has been linked to colon adenocarcinoma in families with familial polyposis. In addition, it enhances the activity of plasminogen activator (PA), which adds to its significance in regulating growth and differentiation.

Background

Developmentally regulated, secreted growth factor homologous to pleiotrophin (PTN), which has heparin binding activity. Binds anaplastic lymphoma kinase (ALK) which induces ALK activation and subsequent phosphorylation of the insulin receptor substrate (IRS1), followed by the activation of mitogen-activated protein kinase (MAPK) and PI3-kinase, and the induction of cell proliferation. Involved in neointima formation after arterial injury, possibly by mediating leukocyte recruitment. Also involved in early fetal adrenal gland development (By similarity).
