Description | This protein, known to retain NFE2L2/NRF2 in the cytosol, also acts as a substrate adapter for the E3 ubiquitin ligase complex formed by CUL3 and RBX1. Its role is to direct NFE2L2/NRF2 towards ubiquitination and subsequently, degradation by the proteasome. As a result of this process, the transcriptional activity of NFE2L2/NRF2 is suppressed, leading to the repression of antioxidant response element-mediated detoxifying enzyme gene expression. |
| Other names | INrf2, also known as Keap1 or Kelch-like ECH-associated protein 1, is a cytosolic inhibitor of Nrf2. It belongs to the Kelch-like family and functions as a molecular adaptor for the ubiquitin ligase complex that targets Nrf2 for degradation. This regulation of Nrf2 activity is crucial for maintaining the cellular redox balance and protecting against oxidative stress. Mutations in the KEAP1 gene, which encodes INrf2, have been associated with various diseases such as cancer and neurodegenerative disorders. The study of INrf2 and its role in Nrf2 regulation has important implications for understanding cellular mechanisms involved in disease progression and therapeutic interventions. |

| Specificity | The Keap1 Antibody is designed to recognize and measure the expression levels of total Keap1 protein in cells. It is capable of detecting the endogenous (naturally occurring) levels of Keap1 within the biological system. This antibody provides a valuable tool for studying the regulation and functions of Keap1 in various cellular processes. |

Target | |
Background | Acts as a substrate adapter protein for the E3 ubiquitin ligase complex formed by CUL3 and RBX1 and targets NFE2L2/NRF2 for ubiquitination and degradation by the proteasome, thus resulting in the suppression of its transcriptional activity and the repression of antioxidant response element-mediated detoxifying enzyme gene expression. Retains NFE2L2/NRF2 and may also retain BPTF in the cytosol. Targets PGAM5 for ubiquitination and degradation by the proteasome. |
| Tissue specificity | Broadly expressed, with highest levels in skeletal muscle. |
| Posttranslational modification | Ubiquitinated by the E3 ubiquitin ligase complex formed by CUL3 and RBX1 and is subject to proteasomal-independent degradation. Quinone-induced oxidative stress, but not sulforaphane, increases its ubiquitination. Ubiquitination and subsequent degradation is most pronounced following prolonged exposure of cells to oxidative stress, particularly in glutathione-deficient cells that are highly susceptible to oxidative stress. |
