Description

The family of transcription factors known as interferon regulatory factors (IRFs) play a pivotal role in regulating interferon expression and IFN-inducible genes when the host encounters viral infections. IRFs are also involved in the defense against pathogens, lymphocyte development, autoimmunity, cell growth, and susceptibility to transformation. IRFs comprise nine members, namely IRF-1, IRF-2, ISGF3γ/p48, IRF-3, IRF-4 (Pip/LSIRF/ICSAT), IRF-5, IRF-6, IRF-7, and IRF-8/ICSBP, and they all possess a similar DNA-binding motif in their amino-terminal domain. These proteins associate in transcription regulation through interactions with proteins that share DNA-binding motifs. IRF-5 is highly expressed in lymphoid tissues and peripheral blood lymphocytes and is vital in the induction of type I interferon genes upon viral infection. Adversely, the IRF-5 signaling pathway activates by different TLRs, including TLR7 and MyD88. Genetic IRF-5 variants have been linked with immune disorders consequent to IFN pathway hyperactivity, including systemic lupus erythematosus.
