Description

IL-17A is a pro-inflammatory cytokine produced by a distinct CD4+ T cell lineage known as Th17 cells. This cystine-linked homodimeric cytokine has been shown to stimulate the production of other pro-inflammatory cytokines like TNF-α, IL-1β, and IL-6. IL-17A also plays a vital role in innate immunity by inducing the production of neutrophil chemoattractants like CXCL1, CXCL6, and IL-8. Interestingly, this cytokine is also involved in mucosal immunity particularly against bacterial infections, but it has also been implicated in some autoimmune disorders.
IL-17A's effects are primarily exerted through its binding to the IL-17RA receptor. This binding triggers the activation of several pathways including the Erk1/2 MAP kinase, PI3K/Akt, p38, and NF-κB pathways. Activated pathways lead to the production of an array of cytokines, chemokines, and other proteins, thus playing a significant role in immune regulation. Some Jaks and Stats have also been observed to undergo phosphorylation in response to IL-17A signaling.
