The inactive NF-κB/Rel transcription factors are bound to IκB proteins within the cytosol (1-3). The majority of stimuli that activate NF-κB initiate a common pathway involving proteasome-mediated degradation of IκB by phosphorylation (3-7). This pathway's critical regulation step involves the activation of the IKK complex, a high molecular weight kinase complex composed of three tightly associated IKK subunits. IKKα, IKKβ, and IKKγ serve as the catalytic and regulatory subunits of the kinase, respectively (8,9). IKK activation is prompted by the phosphorylation of Ser177 and Ser181 in the activation loop of IKKβ (or Ser176 and Ser180 in IKKα). Consequently, the phosphorylation causes structural changes resulting in kinase activation (10-13).
