Description

The inhibitory protein, Id2, is a member of the helix-loop-helix (HLH) protein family which inhibits transcription factors by preventing their DNA binding and dimerization abilities (1-3). Id2 interacts with different transcription factors such as E proteins (5), TCS (4), Pax (6) and the tumor suppressor Rb (1). Its functions in regulating differentiation, proliferation, development as well as tumorigenesis have been widely proven (7-9). Increase in levels of Id2 expression in tumor cells functionally inactivates Rb, leading to cellular transformation and cancer (10,11). The potential therapeutic application of Id2 in cancer treatment has been suggested (12).

Other namesID2, also known as Inhibitor of Differentiation 2, is a protein involved in cell growth inhibition. It belongs to the class B basic helix-loop-helix (bHLH) protein family and is encoded by the bHLHb26 gene. ID2 acts as a DNA binding protein inhibitor, specifically binding to DNA and blocking its binding with other proteins.
GIG8, or Growth Inhibiting Gene 8, is another name for ID2. It is a helix-loop-helix protein that plays a role in regulating cell growth. ID2A and ID2H are alternative forms of the ID2 protein, and MGC26389 is a gene symbol associated with ID2.
In summary, ID2 is a class B bHLH protein that acts as an inhibitor of DNA binding, thus inhibiting cell growth. It is also known as GIG8 and has alternative forms named ID2A and ID2H.
SourceRabbit
SpecificityTotal ID2 can be detected by the ID2 Antibody, which is capable of recognizing the endogenous levels of this protein. The antibody specifically binds to ID2 and can accurately identify the presence of this molecule in various biological samples. Its ability to detect total ID2 makes it a valuable tool for researchers studying the functions of this protein in different physiological processes.

ApplicationFor Western Blotting, the suggested dilutions are between 1:500 to 1:2000, while for Immunohistochemistry, it is recommended to dilute between 1:50 to 1:200. If you plan to use ELISA with peptides, the dilution range is from 1:20000 to 1:40000. It is important to keep in mind these guidelines while performing the experiments to obtain accurate results.

Target

Background

Transcriptional regulator (lacking a basic DNA binding domain) which negatively regulates the basic helix-loop-helix (bHLH) transcription factors by forming heterodimers and inhibiting their DNA binding and transcriptional activity. Implicated in regulating a variety of cellular processes, including cellular growth, senescence, differentiation, apoptosis, angiogenesis, and neoplastic transformation. Inhibits skeletal muscle and cardiac myocyte differentiation. Regulates the circadian clock by repressing the transcriptional activator activity of the CLOCK-ARNTL/BMAL1 heterodimer. Restricts the CLOCK and ARNTL/BMAL1 localization to the cytoplasm. Plays a role in both the input and output pathways of the circadian clock: in the input component, is involved in modulating the magnitude of photic entrainment and in the output component, contributes to the regulation of a variety of liver clock-controlled genes involved in lipid metabolism.

Tissue specificityHighly expressed in early fetal tissues, including those of the central nervous system.
