Description

HMG-CoA reductase is the key enzyme responsible for the biosynthesis of cholesterol and is regulated through negative feedback by sterol and non-sterol metabolites derived from mevalonate, which is the product of the reaction catalyzed by the reductase. In mammalian cells, cholesterol inhibits this enzyme through the internalization and degradation of low density lipoprotein (LDL) via the LDL receptor. Competitive inhibitors of the reductase promote the expression of LDL receptors in the liver, increasing the catabolism of plasma LDL and decreasing the plasma concentration of cholesterol, a critical determinant of atherosclerosis. Multiple isoforms of this gene have been identified via alternatively spliced transcript variants. [provided by RefSeq, Aug 2008].

Other namesThe enzyme known as 3-hydroxy-3-methylglutaryl CoA reductase is also referred to as HMG CoA reductase, HMG CoAR, Hmgcr, and 3H3M. It plays an important role in the biosynthesis of cholesterol by reducing 3-hydroxy-3-methylglutaryl CoA to mevalonate. The enzyme requires NADPH as a cofactor to carry out this reaction. It is a target for drugs such as statins, which are used to lower cholesterol levels in the body. In humans, the HMG-CoA reductase gene is located on chromosome 5. In addition, this enzyme is also known as hydroxymethylglutaryl CoA reductase or 3-hydroxy-3-methylglutaryl-coenzyme A reductase. LDLCQ3 and MGC103269 are alternate names for this gene.
