Description

The Mediator complex plays an essential role in regulated gene transcription of almost all RNA polymerase II-dependent genes. Acting as a bridge between gene-specific regulatory proteins and the basic RNA polymerase II transcription machinery, the Mediator is critical in conveying relevant information to execute transcriptional processes. Upon recruitment to promoters by direct interaction with regulatory proteins, the Mediator functions as a scaffold in assembling a proper preinitiation complex with RNA polymerase II and the general transcription factors. Additionally, it helps inhibit the formation of a transcription initiation complex and down-regulates the TFIIH complex and transcriptional repression by phosphorylating the CTD of the large subunit of RNA polymerase II (RNAp II) and CCNH, respectively. Furthermore, it can act through interaction with MAML1 to hyperphosphorylate the intracellular domain of NOTCH, which leads to its eventual degradation.

Other namesCDK8 is a protein kinase that plays a crucial role in cell division. Also known as Cell Division Kinase 8, this enzyme is responsible for mediating RNA polymerase II transcription. Cyclin-dependent kinase 8, another name for CDK8, is encoded by the CDK8 gene in humans. K35 is a common abbreviation for this protein kinase. The Mediator complex subunit CDK8 is also referred to as Mediator of RNA polymerase II transcription subunit CDK8. This enzyme is essential for regulating different cellular processes, including gene expression and proteins' production. Proteins that work with CDK8 help to keep cell growth in check and prevent unwanted cell division.

ApplicationFor Western blotting, it is suggested to use a dilution of 1:500 to 1:2000. For immunofluorescence or immunocytochemistry, a dilution of 1:100 to 1:500 is recommended. When performing ELISA using peptides, a dilution of 1:20000 to 1:40000 is recommended. To ensure accurate and reliable results, it is important to follow the recommended dilution ratios for each application. These dilution ratios have been optimized for optimal sensitivity and specificity in detecting target proteins or peptides.

Target

Background

Component of the Mediator complex, a coactivator involved in regulated gene transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors. Phosphorylates the CTD (C-terminal domain) of the large subunit of RNA polymerase II (RNAp II), which may inhibit the formation of a transcription initiation complex. Phosphorylates CCNH leading to down-regulation of the TFIIH complex and transcriptional repression. Recruited through interaction with MAML1 to hyperphosphorylate the intracellular domain of NOTCH, leading to its degradation.
