Description | The Mediator complex plays an essential role in regulated gene transcription of almost all RNA polymerase II-dependent genes. Acting as a bridge between gene-specific regulatory proteins and the basic RNA polymerase II transcription machinery, the Mediator is critical in conveying relevant information to execute transcriptional processes. Upon recruitment to promoters by direct interaction with regulatory proteins, the Mediator functions as a scaffold in assembling a proper preinitiation complex with RNA polymerase II and the general transcription factors. Additionally, it helps inhibit the formation of a transcription initiation complex and down-regulates the TFIIH complex and transcriptional repression by phosphorylating the CTD of the large subunit of RNA polymerase II (RNAp II) and CCNH, respectively. Furthermore, it can act through interaction with MAML1 to hyperphosphorylate the intracellular domain of NOTCH, which leads to its eventual degradation. |
