Description | Caspase-9, also known as ICE-LAP6 or Mch6, is a significant member of the caspase family, which consists of cysteine aspartic acid proteases. It plays a critical role in apoptosis (1,2). When cells undergo apoptotic stimulation, cytochrome c is released from the mitochondria and combines with the 47 kDa procaspase-9/Apaf 1. This leads to the activation of caspase-9 mediated by Apaf-1 (3-6). During this process, caspase-9 undergoes intrinsic proteolytic processing, resulting in cleavage at Asp315. This produces a p35 subunit. Additionally, another cleavage occurs at Asp330, generating a p37 subunit. The presence of the p37 subunit can enhance the apoptotic response, amplifying the overall effect (3-6). Subsequently, the cleaved caspase-9 further processes other caspase members such as caspase-3 and caspase-7. This initiates a caspase cascade, ultimately leading to apoptosis (7-10). |

It is regulated by various mechanisms, including phosphorylation and interaction with other proteins, such as protein phosphatase 1, regulatory subunit 56 (PPP1R56). Moreover, dominant-negative mutants of caspase 9 have been generated to investigate the role of caspase 9 in various disease models.
