Description

Interleukin-1 converting enzyme, also known as caspase-1, is a type I cysteine protease enzyme. This class of enzymes also includes caspases -4, -5, -11, and -12. Caspase-1 is responsible for cleaving inflammatory cytokines such as pro-IL-1 and interferon-γ inducing factor (IL-18) into their mature forms. The process involves proteolytic activation of the proenzyme, leading to the formation of a tetramer of active subunits p20 and p10. Caspase-1 has a large pro-domain containing a caspase recruitment domain (CARD). Overexpression of caspase-1 can cause apoptosis.
Deficiency of caspase-1 in mice does not affect apoptosis but does impair the maturation of pro-IL-1β and renders the animals resistant to endotoxic shock. There are at least six identified isoforms of caspase-1, including caspase-1 α, β, γ, δ, ε and ζ. Most isoforms induce apoptosis upon over-expression, with products ranging between 30-48 kDa. The ε isoform of caspase-1 only contains the p10 subunit and acts as a dominant negative without inducing apoptosis. The ζ isoform lacks 39 amino-terminal residues found in the α isoform but induces apoptosis.
Activation of caspase-1 involves the formation of an inflammasome molecular platform that includes caspase-5, Pycard/Asc, and NALP1. This oligomerization process results in the activation of caspase-1 for its cytokine cleavage function.
