| Other names | The protein known as Ataxia telangiectasia and Rad3-related (ATR) plays a crucial role in cellular processes such as DNA repair and cell cycle regulation. It is also referred to as ATR_HUMAN, FCTCS, FRAP Related Protein 1 (FRP1), MEC1, and RAC3. ATR is a serine/threonine protein kinase that functions similarly to the mitosis entry checkpoint 1 homolog (MEC1). Its primary function is to ensure genomic stability by detecting and repairing DNA damage. ATR is involved in signaling pathways that are activated in response to various types of DNA damage, including breaks, cross-links, and replication stress. Once activated, ATR initiates a cascade of phosphorylation events, leading to cell cycle arrest and DNA repair. It also phosphorylates a wide range of downstream targets involved in DNA replication, cell cycle progression, and apoptosis. Defects in the ATR gene can result in a rare genetic disorder called Ataxia telangiectasia and Rad3 related (ATR syndrome) or Seckel syndrome (SCKL). These syndromes are characterized by symptoms such as progressive neurological dysfunction, immune system abnormalities, sensitivity to radiation, and an increased risk of cancer. Research on ATR and its related proteins is crucial for understanding the molecular mechanisms underlying these disorders and developing new therapeutic strategies. |
| Source | Rabbit |
| Specificity | The phospho-ATR(S428) Antibody specifically recognizes the presence of phosphorylated ATR at the S428 site in endogenous levels. This antibody exhibits high specificity towards the phosphorylated form of ATR at S428. |
| Species Reactivity | Human;Mouse;Rat |
| Antigen | phospho -ATR |
| Application | The suggested dilutions for western blot (WB) range from 1:1000 to 1:3000. Immunohistochemistry (IHC) should be diluted at 1:200, while the peptide used in enzyme-linked immunosorbent assay (ELISA) requires dilutions ranging from 1:20000 to 1:40000. |
| Immunogen | Phospho-ATR(S428) - a human-derived peptide that has been chemically synthesized. |
| MW | 300 kDa |
| Properties | |
Concentration | 1mg/ml |
| purification | The purified rabbit serum was used to derive the antibody, which went through affinity purification via both phospho- and non-phospho-peptide affinity columns. This process ensured the removal of unwanted substances and the isolation of the desired antibody. |
| Clonality | Polyclonal Antibody |
| Stability & Storage | To maintain the integrity of the product, it is recommended to store it at a temperature of -20 °C for up to one year. It is important to avoid subjecting the product to multiple freeze/thaw cycles, as this may compromise its quality. |
| Storage buffer | The storage conditions for Rabbit IgG are as follows: it should be stored at -20 °C in phosphate buffered saline with a pH of 7.4. The solution should contain 150mM NaCl, 0.02% sodium azide, and 50% glycerol. It is important to note that the stability of the IgG is guaranteed for a period of 12 months from the date of receipt. |

Background | Serine/threonine protein kinase which activates checkpoint signaling upon genotoxic stresses such as ionizing radiation (IR), ultraviolet light (UV), or DNA replication stalling, thereby acting as a DNA damage sensor. Recognizes the substrate consensus sequence [ST]-Q. Phosphorylates BRCA1, CHEK1, MCM2, RAD17, RPA2, SMC1 and p53/TP53, which collectively inhibit DNA replication and mitosis and promote DNA repair, recombination and apoptosis. Phosphorylates 'Ser-139' of histone variant H2AX/H2AFX at sites of DNA damage, thereby regulating DNA damage response mechanism. Required for FANCD2 ubiquitination. Critical for maintenance of fragile site stability and efficient regulation of centrosome duplication. |
| Tissue specificity | Ubiquitous, with highest expression in testis. Isoform 2 is found in pancreas, placenta and liver but not in heart, testis and ovary. |
| Posttranslational modification | Phosphorylated; autophosphorylates in vitro. |
| Celluar localization | Golgi apparatus;Nucleus; |
| UniPort | Q13535 |
