| Description | Paraformaldehyde, formaldehyde, and other aldehyde reagents are often used to fix cells or tissues. However, one drawback of this fixation process is the formation of cross-links between proteins. These cross-links can interfere with the binding of antibodies to their target antigens, leading to a weakened staining signal during immunostaining. In some cases, false positive staining results may also be observed. It is important to be aware of this potential issue and consider alternative fixation methods or antigen retrieval techniques to minimize the impact on immunostaining outcomes. |
| Usage | To use the rapid antigen repair solution for frozen sections (5×), it is important to prepare it correctly beforehand. This involves diluting the antigen repair solution (1×) with distilled water or MilliQ water for 5 times. In addition, if the rapid antigen repair solution (5×) of frozen sections is stored at 2~8℃, there may be precipitation when it is taken out. If this occurs, simply incubate the reagent at 37℃ and shake it to dissolve the precipitated reagent again. When using the antigen repair solution for paraffin sections, start by dewaxing the sections in xylene for 2×5min, anhydrous ethanol for 2×5min, 90% ethanol for 2×5min, 70% ethanol for 5min, and distilled water 2×5min. Then, soak the sections in antigen repair solution (1×) and incubate at room temperature for 5 min. Make sure to wash the sections 3-5 times with immunostaining solution, 3-5min each time, to remove any residual reagent such as SDS. After this, proceed with subsequent immunostaining steps such as blocking. For frozen sections, wash the sections with immunostaining solution for 5min and then immerse them in antigen repair solution (1×) at room temperature for about 5min. Ensure to wash the sections several times with immunostaining solution to fully remove the remaining reagent such as SDS. Once complete, move on to subsequent immunostaining steps such as blocking. If antigen repair is needed for other types of samples, refer to the steps outlined for paraffin or frozen sections. Always ensure to properly prepare and dilute the antigen repair solution, and wash the sections thoroughly to achieve accurate and reliable results. |
