| Description | Eosin Y, known as the most commonly used cytoplasmic stain, exhibits its versatility when combined with the basic blue dye. This powerful combination, when mixed with hematoxylin, forms the renowned "H&E" dye solution. Notably, Eosin Y functions as an acid stain solution that interacts specifically with proteins abundant in basic amino acids within cells. As a result, the dye protein complex is distinguished by its vivid pink cytoplasmic staining, a characteristic trait that aids in cellular analysis and identification. |
| Usage | Here is a similar content: To start the experimental procedure, we must first deparaffinize and fix the frozen sections. Next, we proceed with Hematoxylin staining and tap water rinsing. In case the regression hematoxylin staining solution is used, it needs to be differentiated and rinsed with tap water. After that, we recommend using an alkaline solution or Scott's Tap Water Substitute for blueing. Rinse again with tap water. Then, we rinse the slide with 95% ethanol for 30 seconds. For step 8, we suggest an acid modified Eosin Y dye solution (alcohol-soluble) by adding 0.5ml glacial acetic acid to 100ml dye solution. For the next step, 9, we use acid modified Eosin Red Y dye solution for counterstaining and let it stay for 30 seconds to three minutes. Lastly, we proceed with dehydration, transparency, and mounting. As a result, the cytoplasm is stained pink to red, while the nuclei are stained blue to blue-black, depending on the hematoxylin stain used. |
