This finding showcases the potential benefits of utilizing insulin-transferrin-selenium as a supplement in cell culture experiments, reducing costs and FBS usage without compromising the quality of cell growth.To prepare the Earle's Balanced Salt Solution (EBSS) according to ITS (Insulin-Transferrin-Selenium) Earle's equilibrium salt solution, follow these steps:
1. Gather the required ingredients for the EBSS preparation.
2. Ensure you have the necessary equipment such as a sterile container, a magnetic stirrer, and a pH meter.
3. Begin by measuring distilled water to meet the desired volume of the EBSS solution. It should be prepared in a sterile environment to prevent any contamination.
4. Transfer the measured distilled water into the sterile container.
5. Add the individual salts and components in the following order: sodium chloride (NaCl), potassium chloride (KCl), calcium chloride (CaCl2), magnesium chloride (MgCl2), sodium bicarbonate (NaHCO3), sodium phosphate dibasic (Na2HPO4), and glucose.
6. Use a magnetic stirrer to mix the solution thoroughly until all the components are dissolved completely.
7. Check the pH of the solution using a pH meter. Adjust the pH if necessary by adding small amounts of hydrochloric acid (HCl) or sodium hydroxide (NaOH) while monitoring the pH level until it reaches the desired range (usually between pH 7.2 to 7.4).
8. Once the pH is within the desired range, check the overall volume of the solution again. Adjust it if needed to meet the desired final volume.
9. Sterilize the EBSS solution using an appropriate method such as autoclaving or filtration, depending on the requirements of your experiment.
