Description

Gill's I stain is considered a progressive cytological stain, while Gill's II and III stains can be either progressive or regressive, depending on the duration of the staining process. The binding occurs between the positively charged aluminum-oxyhematoxylin complex and the negatively charged phosphate group present in the nuclear DNA. As a result of hematoxylin staining, a blue-purple color can be achieved.

Usage
To prepare frozen sections for analysis, follow these steps:
1. Start by dewaxing and hydrating the sections.
2. Next, immerse the sections in Gill's III hematoxylin solution for a duration of 1.5 to 3 minutes.
3. Rinse the sections under running water to remove any excess dye.
4. Apply a differentiation solution to the sections for a period of 20-60 seconds to enhance contrast.
5. Rinse the sections again under running water.
6. Submerge the sections in Scott's Tap Water Substitute or warm slightly alkaline tap water, or use diluted ammonia, for 15 to 60 seconds to further adjust the pH.
7. Rinse the sections once more under running water.
If you are conducting histological analysis, proceed with the following steps:
1. Restain the sections with Eosin Y (either alcohol-soluble or water-soluble) for a duration of 30-60 seconds.
2. Dehydrate the sections, make them transparent, seal them onto a slide, and prepare them for microscopic examination.
If you are performing cytological analysis, follow these steps instead:
1. Restain the sections with either Pasteurized solution OG or EA (ST017) for 1-3 minutes.
2. Proceed with the steps of dehydration, transparency, tablet sealing, and microscopic examination.
By adhering to these guidelines, you can effectively process frozen sections for histological or cytological analysis.
