| Description | Gill's I is a type of cytological stain that is considered as being progressive. On the other hand, Gill's II and III can function either progressively or regressively, depending on the duration of the staining process. The complex formed by the positively charged aluminum-oxyhematoxylin binds to the DNA's negatively charged phosphate group, resulting in a blue-purple coloration. Hematoxylin staining, therefore, has the potential to produce shades of blue and purple. |
| Usage | The process for preparing and staining a smear is as follows: 1. Immerse the smear in 95% ethanol for 15 minutes to fix it. 2. Gently rinse the smear with running water for 30 seconds. 3. Stain the smear with Gill's I hematoxylin solution for a duration of 1.5 to 3 minutes. 4. Rinse the smear with running water to remove excess stain. 5. Submerge the smear in either Scott's Tap Water Substitute (BF058), warm slightly alkaline tap water, or diluted ammonia for 15 to 60 seconds. 6. Rinse the smear again with running water. 7. Dip the smear in 95% denatured alcohol (BF057) or 95% ethanol for 10 repetitions. 8. Reapply the Pasteurized solution OG (ST017) as a redye for 1.5 minutes. 9. Repeat step 7 by dipping the smear in 95% denatured alcohol or ethanol 10 times. 10. Apply Pasteurized dye EA (ST017) as a second redye for 2.5 to 3 minutes. Repeat the dipping process in 95% denatured alcohol or ethanol twice. 11. Rinse the smear in anhydrous ethanol for 1 minute, repeating the step twice. 12. Rinse the smear in xylene or a xylene substitute for 2 minutes, repeating the step twice. 13. Finally, examine the smear under a microscope after applying a cover.
