Testing Principle：

This kit relies on the highly effective double antibody sandwich ELISA technique. It comes with pre-installed anti-human CXCL13 Capture Antibody on a conjugate plate designed for high affinity interactions. The standard substance, sample to be tested and biotin-labeled detection antibody are added sequentially into the HRP-plate well. This initiates a complete oscillation and mixing process, enabling the CXCL13 in the sample to undergo a 2-hour incubation phase at normal room temperature. During this stage, the CXCL13 in the sample combines with the solid phase and detection antibodies, producing reliable and accurate results. After washing thoroughly to remove unbound elements, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added. A further washing stage follows, after which TMB color substrate is added and incubated in the dark at room temperature for color development. The depth of color reaction is in direct proportion to the concentration of CXCL13 in the sample. The termination solution is finally added to stop the reaction, and the absorbance value is taken at 450 nm detection wavelength (correction wavelength 570-630 nm) using a microplate analyzer.

Background：

IL-22 belongs to the IL-10 superfamily, which comprises cytokines involved in cellular inflammatory responses, making them effective regulators. It shares signaling pathways with other family members such as IL-10, IL-26, IL-28A/B, and IL-29 through IL-10R2. This cytokine is produced by activated DCs and T cells to initiate innate immune responses against pathogenic bacteria, with a particular focus on those originating from epithelial cells like those found in the respiratory and intestinal systems. Notably, psoriasis patients show significantly elevated levels of IL-22 in their plasma, which corresponds to the severity of the disease. Additionally, IL-22 serves as a protective factor for hepatocytes, safeguarding against T cell-mediated hepatitis induced by concanavalin A (Con A). Unlike IFN-γ and IL-17, patients with Crohn's disease exhibit high concentrations of IL-22 in their bloodstream.
