Overview

Description

RANTES is a chemoattractant that is known to attract various types of cells such as blood monocytes, memory T-helper cells, and eosinophils. It also stimulates the release of histamine from basophils and activates eosinophils. RANTES primarily binds to CCR1, CCR3, CCR4 and CCR5 chemokine receptors. Interestingly, RANTES is one of the major HIV-suppressive factors produced by CD8+ T-cells. Research has demonstrated that recombinant RANTES protein can inhibit different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV) in a dose-dependent manner.

Other names

RANTES, also known as Beta chemokine RANTES, is a chemokine protein that plays a crucial role in immune responses. It is encoded by the gene CCL5 and acts as a chemoattractant for various immune cells, including eosinophils and T cells. RANTES is produced as a precursor molecule and undergoes processing to generate its active form.
Alternative names for RANTES include C C motif chemokine 5, CCL 5, CCL5_HUMAN, Chemokine (C C motif) ligand 5, and EoCP. It is also referred to as Small inducible cytokine A5 (RANTES). The protein is involved in regulating immune cell migration and activation.
RANTES is secreted by various cell types upon activation and plays a vital role in inflammation and immune responses. It is known to attract eosinophils, which are involved in allergic reactions and asthma. Additionally, RANTES recruits T cells to sites of inflammation and is important for their localization and activation.
The gene CCL5 is located on chromosome 17 and has been associated with various diseases and conditions, including viral infections, autoimmune disorders, and cancer. It is regulated upon activation and is expressed by different cell types, including leukocytes and endothelial cells.
In summary, RANTES is a chemokine protein with multiple functions in the immune system. Its role in immune cell recruitment and regulation makes it an important target for therapeutic interventions in various diseases.

Specificity

The RANTES Antibody is designed to identify the total amount of RANTES present in a biological sample. This antibody has the ability to detect the endogenous levels of RANTES and provide accurate results. By using this antibody for analysis, scientists can gain insights into the expression levels and distribution patterns of RANTES in different tissues and cell types. With its high specificity and sensitivity, the RANTES Antibody is an invaluable tool for researchers studying the role of RANTES in various biological processes.

Application

For Western blot, dilute the antibody at 1:500-1:2000. For immunohistochemistry, use a dilution of 1:50-1:200. In ELISA assays using peptide antigens, dilute the antibody between 1:20000-1:40000. By rearranging the above information, we can create a similar yet distinct piece of content. It is important to carefully adjust dilutions and concentrations to ensure accurate results in various experimental techniques. When conducting Western blot analysis, dilute the antibody at 1:500-1:2000. For immunohistochemistry, use a dilution of 1:50-1:200. Finally, when performing ELISA assays with peptide antigens, dilute the antibody between 1:20000-1:40000 to achieve optimal results.

Immunogen

One could create a peptide that is synthesized and has its origins in human RANTES. To achieve this, the content must closely resemble the original text and ensure its accuracy.

Stability & Storage

To preserve the quality of the product, it is recommended to store it at a temperature of -20 °C for a period of one year. It is important to avoid subjecting the product to repeated freeze/thaw cycles which may compromise its integrity.

Storage buffer

The recommended storage conditions for Rabbit IgG are as follows: it should be stored at -20°C in phosphate buffered saline (pH 7.4) containing 150mM NaCl, 0.02% sodium azide, and 50% glycerol. The stability of the product is guaranteed for a period of 12 months from the date of receipt.

Target

Background

Chemoattractant for blood monocytes, memory T-helper cells and eosinophils. Causes the release of histamine from basophils and activates eosinophils. May activate several chemokine receptors including CCR1, CCR3, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant RANTES protein induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form RANTES(3-68) acts as a natural chemotaxis inhibitor and is a more potent inhibitor of HIV-1-infection. The second processed form RANTES(4-68) exhibits reduced chemotactic and HIV-suppressive activity compared with RANTES(1-68) and RANTES(3-68) and is generated by an unidentified enzyme associated with monocytes and neutrophils (PubMed:16791620, PubMed:1380064, PubMed:8525373, PubMed:9516414, PubMed:15923218). May also be an agonist of the G protein-coupled receptor GPR75, stimulating inositol trisphosphate production and calcium mobilization through its activation. Together with GPR75, may play a role in neuron survival through activation of a downstream signaling pathway involving the PI3, Akt and MAP kinases. By activating GPR75 may also play a role in insulin secretion by islet cells (PubMed:23979485).

Tissue specificity

Expressed in the follicular fluid (at protein level). T-cell and macrophage specific.

Posttranslational modificationN-terminal processed form RANTES(3-68) is produced by proteolytic cleavage, probably by DPP4, after secretion from peripheral blood leukocytes and cultured sarcoma cells. The identity of the O-linked saccharides at Ser-27 and Ser-28 are not reported in PubMed:1380064. They are assigned by similarity.
