Description | The 9-1-1 cell-cycle checkpoint response complex is an essential component for DNA repair. Its primary role is to be recruited to DNA damages by the replication factor C (RFC) clamp loader complex, enabling it to act as a sliding clamp platform for proteins involved in long-patch base excision repair (LP-BER). To aid in this process, the 9-1-1 complex stimulates DNA polymerase beta (POLB) activity, increasing its affinity for the 3' end of the primer-template. Within this complex, endonuclease FEN1 cleaves substrates with double, nick, or gap flaps of varying sequences and lengths, while DNA ligase I (LIG1) aids in the repair of long-patch base excision substrates. Isoform 1 of this checkpoint response complex possesses additional 3'->5' double stranded DNA exonuclease activity, which enhances its ability to play a role in critical DNA repair processes. |
