Overview

Description

NPM1 is a crucial nucleolar protein that is closely associated with nucleolar ribonucleoprotein structures. It is also known to have the ability to bind single-stranded nucleic acids. One of its primary functions is being a crucial component of template activating factor (TAF)-III. Additionally, it plays a pivotal role in controlling centrosome duplication and may be instrumental in the assembly and transportation of the ribosome. This protein has been found to have two alternatively spliced isoforms, both of which have been studied extensively.

Other names

B23, also known as nucleolar phosphoprotein B23 or nucleolar protein NO38, is a crucial component of the nucleolus. It is encoded by the NPM gene in humans. NPM1 belongs to the nucleophosmin/nucleoplasmin family and is commonly referred to as numatrin. It plays a crucial role in various cellular processes.
Nucleophosmin is a phosphoprotein that primarily localizes to the nucleolus, but it can also translocate to other cellular compartments under certain conditions. It has diverse functions, including ribosome biogenesis, DNA repair, centrosome duplication, and regulation of tumor suppressor proteins.
NPM1 is known for its involvement in various human diseases. Mutations in the NPM1 gene have been associated with acute myeloid leukemia (AML), where a specific mutation leads to the aberrant cytoplasmic localization of nucleophosmin. This mutated form of NPM1 is considered a molecular marker for AML diagnosis and prognosis.
In summary, B23, or nucleolar phosphoprotein B23, also known as NPM1, is a crucial protein involved in nucleolar processes, with implications in various cellular functions and human diseases.

Stability & Storage

To preserve the quality of the product, it is recommended to store it at a temperature of -20 °C for up to one year. It is important to avoid subjecting it to repeated freeze/thaw cycles, which could affect its stability and efficacy. Keeping the product at a stable temperature will help to ensure that it remains effective and reliable for as long as possible. Following these storage guidelines can help to maximize the benefits that this product provides.

Storage buffer

The storage condition for Rabbit IgG is as follows: it is stored at -20 °C in phosphate buffered saline with a pH of 7.4. The solution contains 150mM NaCl, 0.02% sodium azide, and 50% glycerol. It is important to note that the Rabbit IgG is stable for 12 months from the date of receipt.

Target

Background

This protein is involved in a variety of cellular functions, including ribosome biogenesis, centrosome duplication, protein chaperoning, histone assembly, cell proliferation, and regulation of tumor suppressors p53/TP53 and ARF. It is known to bind to ribosomes, facilitating their nuclear export, and is also associated with nucleolar ribonucleoprotein structures and interacts with single-stranded nucleic acids. Additionally, it acts as a chaperone for the core histones H3, H2B, and H4.
The protein also plays a role in DNA repair, as it stimulates the activity of the APEX1 endonuclease on double-stranded DNA containing apurinic/apyrimidinic sites but inhibits its activity on single-stranded RNA containing such sites. It may assist with repairing such sites within nucleoli specifically dedicated to repairing rDNA and removing damaged rRNA molecules.
The protein helps regulate centrosome and centriole duplication, working in concert with BRCA2 and being phosphorylated by PLK2 to trigger centriole replication. It also suppresses apoptosis by interfering with the activation of the EIF2AK2/PKR pathway.
In addition, the protein is involved in enhancing the transcription of MYC target genes when it forms a complex with the MYC protein.

Posttranslational modification

Acetylated at C-terminal lysine residues, thereby increasing affinity to histones.ADP-ribosylated.Phosphorylated at Ser-4 by PLK1 and PLK2. Phosphorylation at Ser-4 by PLK2 in S phase is required for centriole duplication and is sufficient to trigger centriole replication. Phosphorylation at Ser-4 by PLK1 takes place during mitosis. Phosphorylated by CDK2 at Ser-125 and Thr-199. Phosphorylation at Thr-199 may trigger initiation of centrosome duplication. Phosphorylated by CDK1 at Thr-199, Thr-219, Thr-234 and Thr-237 during cell mitosis. When these four sites are phosphorated, RNA-binding activity seem to be abolished. May be phosphorylated at Ser-70 by NEK2. The Thr-199 phosphorylated form has higher affinity for ROCK2. CDK6 triggers Thr-199 phosphorylation when complexed to Kaposi's sarcoma herpesvirus (KSHV) V-cyclin, leading to viral reactivation by reducing viral LANA levels.Sumoylated by ARF.May be ubiquitinated. Ubiquitination leads to proteasomal degradation.
