Description

The NFAT (nuclear factor of activated T cells) protein family, comprising NFAT1 (NFATc2 or NFATp), NFAT2 (NFATc1 or NFATc), NFAT3 (NFATc4), and NFAT4 (NFATc3 or NFATx), consists of transcription factors that work in conjunction with AP-1 (Jun/Fos) to regulate gene transcription for an effective immune response (1,2). In their inactive state, NFAT proteins are phosphorylated and located in the cytoplasm. When calcium levels rise, the calcium/calmodulin-dependent serine phosphatase calcineurin is activated and dephosphorylates NFAT proteins, allowing them to enter the nucleus (2). NFAT signaling ceases when calcium levels drop and NFAT is phosphorylated by kinases like GSK-3 or CK1 (3,4). To inhibit calcineurin and prevent NFAT proteins from translocating to the nucleus, immunosuppressive drugs like cyclosporin A and FK506 are used, which keep NFAT proteins in the cytoplasm (5).
