Description | Integrins play a crucial role in the immune system, primarily in leukocyte trafficking and function. They enable immune cells to attach to endothelial and antigen-presenting cells, participate in cytotoxicity, and facilitate extravasation into tissues. The transduction of signals through integrin adhesion receptors is essential for controlling cell functions that rely on gene expression regulation, including proliferation, differentiation, and survival. Although the intracellular domain of integrins lacks catalytic function, it plays a crucial role in interacting with other transducing molecules to facilitate integrin-mediated signaling. One such molecule is JAB1 (Jun activation domain-binding protein 1), which acts as a coactivator of the c-Jun transcription factor. JAB1 has been identified as a protein that interacts with the cytoplasmic domain of the beta2 subunit of the alphaL/beta2 integrin LFA-1. Deficiencies in leukocyte adhesion molecules, specifically CD11/CD18 (beta2 integrins), can lead to structural defects in the common beta subunit (CD18). These defects prevent the formation of heterodimers and the normal expression of these receptors on the cell surface. Consequently, individuals with these deficiencies are at risk of life-threatening bacterial infections. |
| Other names | The protein known as Integrin beta-2, also referred to as CD18, plays a crucial role in cell adhesion and immune responses. It serves as a subunit of several cell surface glycoproteins, such as LFA-1/CR3/p150, that facilitate interactions between leukocytes, complement receptors, and extracellular matrix proteins. CD18 is also essential for the activation and migration of immune cells, as well as for phagocytosis and cytokine production. Mutations in the ITGB2 gene encoding CD18 can lead to various immunodeficiencies, such as leukocyte adhesion deficiency (LAD), which affects the ability of leukocytes to reach sites of infection or inflammation. CD18 is therefore a promising target for the development of anti-inflammatory and immunomodulatory therapies. |

Background | Integrin alpha-L/beta-2 is a receptor for ICAM1, ICAM2, ICAM3 and ICAM4. Integrins alpha-M/beta-2 and alpha-X/beta-2 are receptors for the iC3b fragment of the third complement component and for fibrinogen. Integrin alpha-X/beta-2 recognizes the sequence G-P-R in fibrinogen alpha-chain. Integrin alpha-M/beta-2 recognizes P1 and P2 peptides of fibrinogen gamma chain. Integrin alpha-M/beta-2 is also a receptor for factor X. Integrin alpha-D/beta-2 is a receptor for ICAM3 and VCAM1. Contributes to natural killer cell cytotoxicity (PubMed:15356110). Involved in leukocyte adhesion and transmigration of leukocytes including T-cells and neutrophils (PubMed:11812992). Triggers neutrophil transmigration during lung injury through PTK2B/PYK2-mediated activation (PubMed:18587400). Integrin alpha-L/beta-2 in association with ICAM3, contributes to apoptotic neutrophil phagocytosis by macrophages (PubMed:23775590). |
| Tissue specificity | Leukocytes. |
| Posttranslational modification | Both Ser-745 and Ser-756 become phosphorylated when T-cells are exposed to phorbol esters (PubMed:11700305). Phosphorylation on Thr-758 (but not on Ser-756) allows interaction with 14-3-3 proteins (PubMed:11700305, PubMed:16301335). |
