The product of this gene is a binding protein that interacts with various molecules, including double-stranded RNA (dsRNA), small noncoding RNAs, and mRNAs. These interactions play a crucial role in regulating gene expression and stabilizing mRNAs. Initially identified as a component of the nuclear factor of activated T-cells (NFAT), this protein serves as a subunit of NFAT, which is essential for T-cell expression of interleukin 2. NFAT consists of two proteins, one weighing 45 kDa and the other 90 kDa, with the larger being the product of this gene. These proteins have been found to influence the movement of nuclear mRNA to the cytoplasm. Suppressing the expression of NF45 or NF90 protein has been shown to decelerate cell growth, potentially by impeding mRNA stabilization. Conversely, another version of this gene, known as NF110, is primarily localized in the nucleus and has only minor effects on cell growth when its levels are reduced. The process of alternative splicing produces multiple transcript variants that encode distinct isoforms of the protein. RNA-binding protein that plays an essential role in the biogenesis of circular RNAs (circRNAs) which are produced by back-splicing circularization of pre-mRNAs. Within the nucleus, promotes circRNAs processing by stabilizing the regulatory elements residing in the flanking introns of the circularized exons. Plays thereby a role in the back-splicing of a subset of circRNAs (PubMed:28625552). As a consequence, participates in a wide range of transcriptional and post-transcriptional processes. Upon viral infection, ILF3 accumulates in the cytoplasm and participates in the innate antiviral response (PubMed:21123651). Mechanistically, ILF3 becomes phosphorylated and activated by the double-stranded RNA-activated protein kinase/PKR which releases ILF3 from cellular mature circRNAs. In turn, unbound ILF3 molecules are able to interact with and thus inhibit viral mRNAs (PubMed:21123651, PubMed:28625552).
