Overview

Description

Chromosome segregation heavily relies on the crucial involvement of HEC1, which effectively interacts with multiple proteins involved in regulating the G2/M phase. The coiled-coil domains of HEC1 serve as the interface for these interactions. To ensure accurate chromosome segregation, it is imperative that Nek2 phosphorylates HEC1. This phosphorylation event is vital for maintaining the fidelity of chromosome segregation. Additionally, HEC1 plays a crucial role in the recruitment of both Mps1 kinase and Mad1/Mad2 complexes to kinetochores. These interactions are essential for the proper functioning of the cell division process.

Other names

HEC, also known as Highly Expressed in Cancer, is a protein that is commonly found in cancerous cells. It is characterized by its rich leucine heptad repeats and is also known by other names such as Highly Expressed in Cancer Protein and Kinetochore-associated Protein 2. HEC is encoded by the KNTC2 gene and is responsible for binding to and stabilizing the kinetochore, a protein complex involved in cell division. It is also known as HsHec1 or hsNDC80, and is homologous to the NDC80 protein found in yeasts. NDC80 is crucial for the correct alignment of chromosomes during cell division, and mutations in the gene encoding NDC80 can lead to abnormal cell division and the development of cancer. Retinoblastoma-associated protein HEC (TID3) has also been linked to HEC and its role in cell division. Overall, HEC is an important protein in maintaining normal cell division and preventing the development of cancer.

Specificity

The HEC1 antibody is capable of recognizing the complete amount of HEC1 present within an organism. This particular antibody effectively targets the endogenous levels of HEC1, thereby providing a sound basis for research in a variety of scientific fields. By efficiently detecting HEC1 at its total concentration, the HEC1 antibody can serve as a useful tool in the study of this protein's role in various physiological processes.

Application

ELISA (peptide) is performed with a dilution range of 1:20000-1:40000. In Western blotting, the recommended dilution range is 1:500~1:3000, while for immunofluorescence (IF) and immunocytochemistry (ICC), the dilution range is 1:100~1:500. It is important to adhere to these dilution guidelines to achieve accurate and reliable results.

Target

Background

The NDC80 protein acts as a crucial component of the NDC80 complex, which is essential for proper chromosome segregation and the spindle checkpoint function. It helps maintain kinetochore integrity and organizes stable microtubule binding sites on the outer plate of the kinetochore, thereby facilitating end-on attachment of kinetochores to spindle microtubules. Additionally, the NDC80 complex collaborates with the SKA1 complex to enhance microtubule affinity and track depolymerizing microtubules. Without the NDC80 protein, chromosome congression is impaired, leading to defects in cell division. Overall, the NDC80 protein is an integral part of the molecular machinery that ensures accurate chromosome segregation during cell division.
